摘要 目的建立高效液相色谱-蒸发光散射检测器(HPLC-ELSD)测定方法,对抗感颗粒进行山银花检查。方法以山银花中两种专属三萜皂苷—灰毡毛忍冬皂苷乙和川续断皂苷乙作为指标成分,采用Thermo Hypersil Gold C_(18)色谱柱(4.6 mm×250 mm,5μm),以水-乙腈为流动相进行梯度洗脱,柱温32℃,体积流量1.0m L·min^(-1),蒸发光散射检测器检测(漂移管温度90℃,雾化器温度60℃,N_2载气流速1.6 L·min^(-1))。结果在收集的53批样品中,49批样品检出了至少一种山银花专属三萜皂苷,存在以山银花投料的情况。结论建立的方法准确、可靠,专属性强,可用于检查抗感颗粒中山银花的使用情况。 Objective To establish an HPLC - ELSD method for the inspection of Shanyinhua( Lonicerae flos)in Kanggan Granules. Methods Macranthoidin B and dipsacoside B,which are both characteristic triterpenoid saponins of Lonicerae flos,were used as the detection targets. The method was carried out on a Thermo Hypersil Gold C18 column(4. 6 mm × 250 mm,5 μm)with the mobile phase consisted of water - acetonitrile by gradient elution. The column temperature was 32 ℃,and the flow rate was 1. 0 mL·min - 1 . The drift tube and atomization temperatures of ELSD were set at 90 ℃and 60 ℃,respectively,with the flow rate of N2 at 1. 6 L·min - 1 . Results 53 batches of samples were measured. At least one characteristic triterpenoid saponin of Lonicerae flos was detected in 49 batches of samples,which indicated Lonicerae flos had been used in corresponding batches of Kanggan Granules. Conclusion The established method was highly accurate, specific,and reliable,which can be applied to detect the use of Lonicerae flos in Kanggan Granules.
