摘要
目的探讨乌司他丁对脓毒血症鼠小肠脂肪组织APN mRNA表达的影响及与TLR4的关系。方法 30只Wistar大鼠随机分为健康对照组(C组,10例)、LPS组(L组,10例)和乌司他丁组(U组,10例),给予乌司他丁48h后处死大鼠,分别于注射前0h、注射后2h、4h、8h、24h、48h检测大鼠血清APN及IL-1、IL-6、TLR4水平,处死大鼠后取小肠脂肪组织检测APN mRNA和TLR4 mRNA的表达情况。结果三组大鼠在注射前0h及注射后2h血清APN水平及IL-1、IL-6、TLR4水平差异无统计学意义(P>0.05);注射后4h、8h、24h、48h,L组与U组大鼠血清APN水平明显低于C组,IL-1、IL-6、TLR4水平明显高于C组,差异有统计学意义(P<0.05);与L组相比,注射后4h、8h、24h、48h,U组大鼠血清APN水平明显高于L组,而IL-1、IL-6、TLR4水平明显低于L组,差异有统计学意义(P<0.05);与C组相比,L组与U组大鼠小肠脂肪组织APN mRNA水平较C组明显降低,L组降低幅度更大,而TLR4 mRNA表达则明显升高,且L组升高幅度更大,差异有统计学意义(P<0.05)。结论乌司他丁可以提升脓毒血症鼠脂联素水平,减少TLR4表达及炎症因子释放,从而减轻炎症反应。
Objective Explore the effect of ulinastatin on the expression of APN mRNA of the adipose tissue in small intes- tine in rats with septicopyemia and it' s correlation with TLR4. Methods 30 Wistar rats were divided into health control group ( group C, 10 cases), LPS group ( group L, 10 cases ) and ulinastatin group ( group U, 10 cases) randomly, injection ulinastatin in rats and 48 hours later killed the rats, test serum APN and the levels of IL-1, IL-6 and TLR4 of rats in 0 hours before injection of and 2h,4h ,8h ,24h and 48h after injection of ulinastatin respectively, take the adipose tissue in small intestine after killed the rats and test the expression of APN mRNA and TLR4 mRNA. Results There are no significant difference in serum APN and the levels of IL-1, IL-6 and TLR4 of rats in 0 hours before injection and 2h after injection of ulinastatin( P 〉 0.05 ). The serum APN levels of rats in group U and group L significantly lower than group C after 4h,8h,24h and 48h of injection ulinastatin, and the levels of IL-1, IL-6 and TLR4 of rats in group U and group L significantly higher than group C, which had a significant difference ( P 〈 0.05 ). The levels of APN mRNA of the adipose tissue in small intestine in rats in group L and group U significantly lower than group C, the group L reduction more, but the expression of TLR4 were significantly rise and group L rise more, it's had a significant difference (P 〈 0.05 ). Conclu- sion Ulinastatin can rise the level of adiponectin in rats with septieopyemia, reduce the expression of TLR4 and the release of inflam- matory factors, thus reduce inflammation.
