蓝莓对非酒精性脂肪肝大鼠Nqo1蛋白表达的影响 被引量:3

Effect of Blueberry Compound on Hepatocyte Expression of Nqo1 in Rats Model of Non-alcoholic Fatty Liver Disease

摘要 目的:探讨蓝莓对非酒精性脂肪肝大鼠的保护作用及机制。方法:40只健康SD大鼠随机均分为正常组、蓝莓组、异甘草酸镁组及模型组,除正常组用普通饲料喂养,其余各组均使用高脂饲料喂养,蓝莓组与异甘草酸镁组在每日固定时间分别灌服蓝莓浆及异甘草酸镁溶液,正常组和模型组灌服等量生理盐水,8周后处死所有大鼠,取肝组织切片油红染色观察各组肝脏脂肪变情况,采用自动生化仪检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、甘油三酯(TG)、总胆固醇(TC)含量;ELISA法检测肝组织肿瘤坏死因子α(TNF-α)、白细胞介素-8(IL-8)水平、超氧化物歧化酶(SOD)活性,Western blot检测大鼠肝组织中醌氧化还原酶1(Nqo1)的表达。结果:正常组大鼠肝组织切片油红染色仅见少量分散脂滴,模型组可见大量红色脂滴,蓝莓组与异甘草酸镁组脂滴明显少于模型组;与蓝莓组及异甘草酸镁组比较,模型组大鼠血清ALT、AST、TG、TC及肝组织TNF-α、IL-8含量明显升高(P<0.05),SOD活性明显降低(P<0.05);相对于正常组,蓝莓组和异甘草酸镁组Nqo1蛋白的表达明显升高(P<0.05)。结论:蓝莓对非酒精性脂肪肝大鼠肝损伤有较好的保护的作用,其机制可能与上调肝组织中Nqo1蛋白的表达有关。 Objective: To investigate the protection effect of blueberry on rats with non-alcoholic fatty liver disease and its mechanism. Methods: Forty healthy SD rats were randomly divided into normal control group,blueberry group,magnesium isoglycyrrhizinate group and model group. The rats in normal control group were given normal diet,while other groups were all given high fat diet. Blueberry group and magnesium isoglycyrrhizinate group were given blueberry juice by intragastric administration while normal group and model group were given equivalent volume of physiological saline by intragastric administration at the same time per day. At the end of 8thweek,all rats were sacrificed. Pathological changes in the hepatic tissue were observed by Oil-Red staining. The contents of alanine aminotransferase( ALT),aspartate aminotransferase( AST),triglyceride( TG) and total cholesterol( TC)in serum were detected by automatic biochemical analyzer. The contents of TNF-α,IL-8 and SOD activity in hepatic tissue were detected by ELISA. The expression of Nqo1 in liver tissue was detected by Western blot. Results: The Oil-Red staining result showed that only a little dispersed lipid droplet could be seen in normal group,blueberry group and magnesium isoglycyrrhizinate group while a lot of dispersed lipid droplet in model group. Compared with blueberry group and magnesium isoglycyrrhizinate group,the contents of serum ALT,AST,TG and TC increased significantly,and the contents of TNF-α and IL-8 in heptic tissue inceased significantly( P〈0. 05) while SOD activity decreased significantly( P〈0. 05). Compared with normal group,the protein expression levels of Nqo1 in blueberry group and magnesium isoglycyrrhizinate group increased significantly( P〈0. 05). Conclusion: Blueberry can play a protective role in liver injury of non-alcoholic fatty liver of rats and the mechanism might be associated with upregulating the expression of Nqo1 in hepatic tissue.
