摘要
目的通过研究慢性宫颈炎、宫颈上皮内瘤样病变和宫颈鳞癌中IKKβ的表达情况,初步探讨IKKβ在宫颈鳞状上皮癌变过程中的作用。方法选取中国医科大学附属盛京医院病理科2010-2015年的100例宫颈组织石蜡标本。所有标本都由两名病理科医师独立诊断并复核确诊,按病理类型分为4组:慢性宫颈炎组(20例),CINⅠ组(20例),CINⅡ~Ⅲ组(20例),宫颈鳞癌组(40例)。采用免疫组织化SP法检测IKKβ在慢性宫颈炎、宫颈上皮内瘤样病变和宫颈鳞癌中的表达,比较各组间IKKβ的平均光密度值。结果 IKKβ阳性表达主要定位于宫颈鳞状上皮细胞的细胞浆,少数可见胞核着色。随着宫颈病变的进展,IKKβ的平均光密度值逐渐增加。宫颈鳞癌组的IKKβ平均光密度值较宫颈炎组、CINⅠ组和CINⅡ~Ⅲ组均显著升高(P〈0.01)、而宫颈炎组和CINⅠ组的平均光密度值比较,差异无统计学意义(P〉0.05)。结论本研究运用免疫组织化学方法检测出人类宫颈鳞状上皮癌变过程中各病变阶段鳞状上皮细胞胞浆中均有IKKβ的表达;且随着宫颈病变的进展,IKKβ的平均光密度值逐渐增加,在宫颈鳞癌组织中呈高表达,表明IKKβ的过度表达在宫颈鳞状上皮癌变道程中起促进作用。
Objective To explore the IKKβ expression in cancerization process of cervical squamous cell: chronic cervicitis,cervical intraepithelial neoplasia and cervical cancer,and to discuss the role of IKKβ in cancerization process of cervical squamous cell. Methods From 2010 to 2015,100 cases of cervical tissue paraffin specimens were selected in our hospital. All specimens were independently diagnosed by two pathologists,and according to the pathological types all specimens were divided into 4 groups: chronic cervicitis group( 20 cases),CINⅠgroup( 20 cases),CINⅡ-Ⅲ group( 20 cases),cervical squamous carcinoma group( 40 cases). Immunohistochemical method was used to detect IKKβ expression in chronic cervicitis,cervical intraepithelial neoplasia lesions and cervical squamous cell carcinomas,to compare the IKKβ average optical density value in each group. Results IKKβ positive expression mainly located in cervical squamous cell cytoplasm,while,a few visible nucleus were stained. Following the progress of the cervical lesions,the IKKβ average optical density value increased. In cervical squamous carcinoma group,IKKβ average optical density value was significantly higher( P 〈 0. 01) than those in cervicitis group,CINⅠgroup and CINⅡ-Ⅲ group; whereas the difference of IKKβ average optical density value between cervicitis group and CINⅠgroup was not statistically diffierent( P 〉 0. 05). Conclusion Positive IKKβ expression was found in the cytoplasma of chronic cervicitis,cervical intraepithelial neoplasia and cervical cancer; and the IKKβ expression level was closely related with the degree of cervical lesions,indicating the IKKβ important role in the cancerization process of cervical squamous cell.
