摘要
目的:考察百事乐加味方对脑卒中后抑郁症(PSD)大鼠海马、前额叶皮层病理形态及脑源性神经影响因子(BDNF)水平的影响。方法:采用大脑中动脉线栓(MCAO)法建立大鼠局灶性脑缺血模型,再结合慢性不可预见的温和性应激(CUMS)和孤养法建立PSD,通过HE染色和免疫组化技术,观察百事乐加味方对PSD大鼠海马、前额叶皮层形态及BDNF的影响。结果:HE染色显示:百事乐加味方组大鼠海马CA1区神经元排列稍不整齐,大部分神经元细胞核、核膜结构较清楚,部分细胞尼氏体消失,少部分细胞胞浆浓缩深染,可见少部分变性或坏死的细胞,损伤程度较PSD组轻;百事乐加味方组大鼠前额叶皮层细胞形态未发生明显改变,结构部分清晰,胞数目减少,内锥体细胞层和多形细胞层损伤较PSD组轻。免疫组化结果显示:百事乐加味方上调PSD大鼠海马、前额叶皮层BDNF的表达(P<0.01)。结论:百事乐加味方增加海马-前额叶皮层环路神经可塑性可能是其治疗PSD的主要机制之一。
Objective: To investigate the effects of the modified Baishile Formula on the morphology of hippocampusprefrontal cortex circuit and the level of BDNF in the rat with PSD model. Methods: PSD mode1 was established with chronic unpredictable mild stress(CUMS) on the basis of focal cerebral ischemia model that induced by the method of middle cerebral artery occlusion(MCAO). HE staining technology was used to observe the changes in the morphology of hippocampus and prefrontal cortex, and the immunohistochemistry technology was used to detect the expression of BDNF. Results: HE staining showed that the neurons of the modified Baishile Formula group were irregularly arranged in CA1 area of hippocampus, where the Nissl body appeared and the cytoplasm was condensed and deeply condensed. But, both of the neuronal nuclei and the nuclear membrane were clearly viewed under the microscope. The injury degree of the modified Baishile Formula group was lower that of PSD group. The cellular morphology of the modified Baishile Formula group was not significantly changed in the prefrontal cortex. The structure was clear and the number was decreased. But, the damage in the internal pyramidal cell layers and polymorphic cell layers in the modified Baishile Formula group were lower than those in PSD group. Immunohistochemical results showed that modified Baishile Formula up-regulated the expression of BDNF in hippocampus and prefrontal cortex of PSD rat(P〈0.01). Conclusion: The modified Baishile Formula has a therapeutic effect on PSD, which might be related to an increase of neuronal plasticity in hippocampus-prefrontal cortex circuit.
