牛蒡根水提物对血管内皮细胞保护作用的研究 被引量:1

Protective effect of water extract of burdock root on vascular endothelial cells

摘要 目的:探讨牛蒡根水提物对血管内皮细胞损伤的保护作用及其机制。方法:用脂多糖(LPS)诱导血管内皮细胞损伤模型,随机分组为空白对照组,模型组和牛蒡根水提物低、中、高浓度组。用酶联免疫吸附试验(ELISA)测定细胞培养上清中人细胞间黏附分子-1(ICAM-1)、人肿瘤坏死因子-α(TNF-α)、人基质金属蛋白酶-1(MMP-1)、人超敏C反应蛋白(hs-CRP)水平;用PCR检测细胞Toll样受体4(TLR4)、核因子(NF-κB)的抑制因子(I-κB)的表达水平。结果:与模型组比较,药物治疗组各组炎性因子ICAM-1、TNF-α、MMP-1及hsCRP水平均降低(P<0.05),TLR4 m RNA表达水平降低(P<0.05),I-κB m RNA表达水平增高(P<0.05)。结论:牛蒡根水提物可能是通过抑制细胞TLR4、NF-κB的表达,从而抑制炎性因子ICAM-1、TNF-α、MMP-1及hs-CRP等的产生继而保护细胞的。 Objective: To explore the protective effect of water extract of burdock root on the vascular endothelial cells and its mechanism. Methods: The endothelial cell injury model was established with lipopolysaccharide(LPS). The cells were randomly divided into the control group, model group, low, middl and high dosage of water extract of burdock root. Enzymelinked immunosorbent assay(ELISA) was used to determine the levels of intercellular adhesion molecule 1(ICAM-1), tumor necrosis factor alpha(TNF-α), matrix metalloproteinase 1(MMP-1), and C-reactive protein concentrations(hs-CRP) in cell culture supernatant. PCR was used to detect the m RNA expression of toll-like receptor 4(TLR4), nuclear factor(NF-κB) inhibitory factor(I-κB) in cells. Results: Compared with the model group, the level of inflammatory factors including ICAM-1, TNF-α, MMP-1, hs-CRP in the drug treatment groups were decreased(P〈0.05), the m RNA expression of TLR4 were decreased(P〈0.05) and the m RNA expression of I-κB were increased(P〈0.05). Conclusion: The water extract of burdock root had a protective effect on the injuried cells, which could be likely to relate to a down-regulation of the expression of TLR4 and NF-κB that could inhibit the production of inflammatory factors such as ICAM-1, TNF-α, MMP-1, and hs-CRP.
