摘要
目的建立HPLC内标手性拆分法分析雨生红球藻片中2种虾青素光学异构体含量的方法。方法采用HPLC法,以反式-β-8'-胡萝卜醛为内标,色谱柱:YMC-Carotenoid TMS(250 mm×4.6 mm,5μm);流动相:甲醇-甲基叔丁醚-1%磷酸溶液,梯度洗脱;流速:1.0 ml·min-1;检测波长:474 nm。结果虾青素异构体在0.5185~5.185μg·ml-1范围内进样量与峰面积线性关系良好(r=0.9999)。平均回收率为99.40%,RSD为4.53%。结论方法简便、快速准确、灵敏度高,重复性好。
Objective To establish a method for chiral separation and determination of astaxanthin stereoisomers in Yushenghongqiuzao tablets using HPLC internal standard method. Methods With trans-β-Apo-8'-carotenal as the internal standard,the HPLC system was performed on a YMC-Carotenoid TMS column( 250 mm × 4. 6mm,5 μm) with methanol-methyl tert-butyl ether( MTB)-1 % phosphoric acid as the mobile phase in a gradient mode at a flow rate of 1. 0 ml·min- 1,detected with UV at 474 nm. Results There was a good linear relationship within the rage 0. 5185- 5. 185 μg·ml- 1of astaxanthin stereoisomers,and the correlation coefficient( r) was0. 9999. The result( n = 6) of the average recovery was 99. 40 %( RSD = 4. 53%). Conclusion This method is simple,quick,accurate and sensitive.
