摘要
目的建立头痛颗粒含量测定方法。方法用HPLC法,采用ZORBAX Eclipse XDB-C18色谱柱(4.6 mm×150 mm,5μm),流动相:甲醇-0.3%冰醋酸溶液(8∶92),检测波长:220 nm,柱温:40℃,流速:0.5 ml·min-1。结果天麻素在0.5~1.5μg范围内与其峰面积有良好的线性关系,r=0.9999,平均回收率为99.92%,RSD为2.19%,3批头痛颗粒中天麻素平均含量为0.5156 mg·g-1。结论本法稳定可靠,可用于头痛颗粒的质量控制。
Objective To establish a quantitative determination method of granules for headache. Methods HPLC was used to determine gastrodin in the granules. The sample was extracted twice with methanol under ultrasonic management for 30 min. Chromatographic conditions consisted of ZORBAX Eclipse XDB-C18 column,a mobile phase composed of methanol and acetic acid,a velocity of 0. 5 ml·min- 1,a detection wavelength of 220 nm,and a column temperature of 40 ℃. Results Within the range of 0. 5- 1. 5 μg,the calibration curve was linear( r = 0. 9999). The mean recovery rate was 99. 92%( RSD2. 19%). The mean content of gastrodin in its preparations of three batches was 0. 5156 mg·g- 1. Conclusion This method is stable and accurate,which can be used as a quality control for granules for headache.
