摘要
目的:分析氯胺酮对SD幼鼠海马GAP-43蛋白表达的影响。方法:取7~10日龄的SD幼鼠36只,随机分为A、B、C三组各12只。A组注射生理盐水50 mg/kg,B组注射氯胺酮25 mg/kg,C组注射氯胺酮50 mg/kg。24 h后处死小鼠取海马组织,分别采用免疫组化与Western Blotting检测海马GAP-43蛋白的表达情况。结果:经图像分析软件处理后得知,A组、B组、C组条带灰度值分别为187.43±7.98、164.87±8.35、154.31±6.98,组间比较差异均有统计学意义(P〈0.01)。A组、B组、C组GAP-43阳性细胞计数分别为(87.45±14.98)个/mm2、(66.48±12.36)个/mm2、(35.32±9.93)个/mm2,组间比较差异均有统计学意义(P〈0.01)。结论:氯胺酮可明显降低SD幼鼠海马GAP-43蛋白的表达水平,这可能与其影响幼鼠神经系统发育有关。
Objective: To study the effect of ketamine on the expression of GAP-43 protein in hippocampus of SD rats. Methods:Thirty six SD rats who were 7 to 10 days old,were randomly divided into three groups. Group A of 12 rats were injected with saline50 mg / kg,group B of 12 rats were injected with ketamine 25 mg / kg,group C of 12 rats were injected with ketamine 50 mg / kg. All rats were sacrificed and the hippocampuses were drawn out after 24 hours. Immunohistochemistry and Western Blotting were used to detect the expressions of hippocampal GAP-43 protein. Results: Then we analyzed the results by image analysis software,gray stripe value of group A was 187.43±7. 98,striped gray value of group B was 164. 87 ± 8. 35,striped gray value of group C was 154. 31 ± 6. 98,there were statistical differences between three groups( P〈0.01). The GAP-43 positive cell count in group A was( 87.45± 14.98) / mm2,group B was( 66.48± 12. 36) / mm2,group C was( 35.32± 9.93) /mm2,there were statistical differences between three groups( P 0. 01).Conclusion: Ketamine can significantly reduce the expression level of GAP-43 protein in the hippocampus of SD rats,which may be related to the development of the nervous system in young rats.
