两种郁金香品种愈伤组织诱导及显微观察 被引量:1

摘要 以海顿和布拉两种郁金香(Tulipa gesneriana)品种的种球为外植体,采用MS培养基附加不同浓度的激素,进行愈伤组织的诱导,并对其愈伤组织进行显微观察。在该过程中,对6-BA、NAA、2,4-D、KT等激素条件对郁金香愈伤组织诱导的影响进行了正交实验分析。结果显示,采用预先冷处理的种球作为外植体,在MS培养基附加1.0 mg/L 6-BA、0.1 mg/L KT和0.5 mg/L 2,4-D中,黑暗培养7 d,后转入光照12 h/d,可诱导出白色、细碎、状态良好的愈伤组织。在显微观察中,发现在该组合下,海顿郁金香的愈伤组织生长较好,内含物颗粒量大,品质较高。 In this article,with bulbs of Brad and Haydn tulip( Tulipa gesneriana) as explants,MS medium plus different concentrations of plant growth regulators was used in callus inducement. The callus was microexamined. In this process,orthogonal experimental methods were used to analysis the impact of 6-BA,NAA,2,4-D and KT to callus inducement of Tulips. The results showed that,after cool pretreatment,the explants of bulbs were cultured on MS medium supplemented with 1. 0mg / L 6-BA,0. 1mg / L KT and 0. 5mg / L 2,4-D. Firstly,the culture was in dark for 7 days,and then transferred to 12 h / d light. Then the white,loose and good status callus was induced.The microexamination results showed that the callus of Haydn tulip grew better,and the cell inclusion particles were bigger and had higher quality in abovementioned conditions.
