夏枯草口服液对雌激素受体阳性乳腺癌细胞凋亡的影响 被引量:11

Apoptotic Effect of the Extract of Xiakucao Oral Solution on Estrogen Receptor-positive Human Breast Cancer Cells

摘要 目的:研究夏枯草口服液对雌激素受体(ER)阳性人乳腺癌细胞凋亡的影响。方法:将ER阳性型人乳腺癌细胞系MCF-7及ER阴性型人乳腺癌细胞系MDA-MB-231、SK-BR-3分别与不同浓度(0、25、50、100 mg/ml)的夏枯草口服液提取物共孵育。采用噻唑蓝(MTT)法检测细胞存活率;流式细胞术及Annexin V/PI双染色检测细胞周期和细胞凋亡;ELISA法检测雌激素应答基因表达量;Western blot法检测ERα蛋白表达情况。结果:1夏枯草口服液提取物呈剂量依赖性地抑制MCF-7细胞的生长,但对MDAMB-231、SK-BR-3细胞无显著作用。2夏枯草口服液提取物可诱导MCF-7细胞产生凋亡,在mRNA水平降低MCF-7细胞内雌激素应答基因的表达,并降低ERα蛋白表达。结论:夏枯草口服液可诱导ER阳性乳腺癌细胞的凋亡,用于治疗ER阳性乳腺癌。 Objective: To study the potential apoptotic effect of Xiakucao oral solution,a patent traditional Chinese medicine,on estrogen receptor( ER) positive human breast cancer cells. Methods: The extract of Xiakucao oral solution( EPV) was treated in ER-positive type human breast cancer cell line MCF-7 and ER-negative type human breast cancer cell line MDA-MB-231,SK-BR-3,respectively. Relative cell viabilities were determined by the MTT method; apoptosis and cell cycle were analyzed by Annexin V / PI dual staining with flow cytometry; the protein expression levels of ERα were detected by Western blot. Gene expression levels were analyzed using ELISA assay kit. Results: 1 EPV reduced the viability of MCF-7cells dose-dependently. However,EPV did not show significantly inhibitory effect on the viabilities of MDA-MB-231 and SKBR-3 cells. 2 EPV induced apoptosis in MCF-7 cells. EPV reduced the level of mRNA within the MCF-7 cell luciferase activity and estrogen-responsive gene expression,and a dose-dependent decrease ERα. Conclusion: EPV could induce apoptosis in ER-positive breast cancer cells,which provide novel information for clinical application of xiakucao oral solution for the ER-positive breast cancer patients.
