聚酰胺树脂分离纯化黑米中矢车菊素-3-葡萄糖苷工艺研究 被引量:6

摘要 通过单因素试验和正交试验,确定了聚酰胺树脂分离纯化黑米中矢车菊素-3-葡萄糖苷的最佳工艺参数。以矢车菊素-3-葡萄糖苷得率为考察指标时,其最佳工艺条件为:上样液pH4、上样液质量浓度25mg/ml、上样液流速6ml/min、洗脱液乙醇体积分数50%、洗脱流速3ml/min;以纯度为考察指标时,该最佳工艺条件为:上样液pH3、上样液质量浓度25mg/ml、上样液流速3ml/min、洗脱液乙醇体积分数30%、洗脱流速6ml/min。聚酰胺树脂能有效纯化黑米花色苷,最终所得样品纯度达到80.84%,大部分黄酮类杂质被有效去除。 Through single factor experiment and orthogonal experiment,the optimal parameter of polyamide resin purification for cyanidin-3-glucoside from black rice was determined.Results showed that:with yield as index,the optimal process conditions were sample solution concentration of 25mg/ml,flow rate of 6ml/min,elution liquid ethanol content of 50%,elution rate of 3ml/min and pH3;with the purity as index,the optimal process conditions:pH3of sample solution,sample solution concentration of 25mg/ml,flow rate of 3ml/min,elution liquid ethanol volume fraction was 30%,elution flow rate increase6ml/min in the.The purification of cyanidin-3-glucoside from black rice by polyamide resin was effective,and the purity of the cyanidin-3-glucoside obtained 80.84%,indicating that most impurity substance such as flavonoids was removed.
