摘要
目的研究茶树菇提取物对小鼠抗疲劳及耐缺氧活性的影响。方法将昆明小鼠随机分为5组,连续灌胃给药28天,观察小鼠力竭跑步时间、疲劳转棒时间、负重游泳时间,以及缺氧和中毒状态下的存活时间;小鼠游泳20min后,测定小鼠肝脏ATP含量、血浆及肝脏中SOD和MDA含量。结果小鼠急性毒性实验结果显示,茶树菇提取物无急性毒性反应。同时,茶树菇提取物可以明显增加小鼠的运动时间,提高缺氧及中毒情况下小鼠存活时间,提高小鼠肝脏ATP含量,提高血浆及肝脏SOD活力,降低MDA含量。结论茶树菇提取物具有显著的抗疲劳及耐氧化活力,其作用可能部分是通过调节能量储备和提高抗氧化酶活性实现的。
Objective The present study aims to investigate the anti-fatigue and anti-hypoxia effects of Agrocybe cylindracea extract in mouse model. Methods KM mice were divided into five groups randomly( n = 16 / group; equal numbers of male and female),and orally treated with saline( 0. 1 ml /20 g serving as vehicle group),0. 6g / kg rhodiola capsule( serving as positive group) and Agrocybe cylindracea extract at doses of 0. 5 g / kg,1. 0 g / kg and 1. 5 g / kg once a day for 28 days. At the end of drug administration,following forced running test,rotating rod test,loading swimming test,normobarie hypoxia test and sodium nitrite toxicosis test were performed. Results Through rotating rod,forced running test,loading swimming test,the anti-fatigue activity of Agrocybe cylindracea extract was demonstrated. Via normobarie hypoxia test,sodium nitrite toxicosis test,Agrocybe cylindracea extract was confirmed to possess anti-hypoxia effect. Agrocybe cylindracea extract treatment significantly enhanced the liver levels of ATP,superoxide dismutase( SOD),and decreased the content of malondialdehyde( MDA). Conclusion We conclude that Agrocybe cylindracea extract efficiently enhances mouse fatigue endurance and anti-hypoxia capability partly through energy reserves and antioxidant enzyme activity.
