头部电针刺激协同神经干细胞移植对脑瘫大鼠脑组织形态学的影响 被引量:2

Effect of Head Electro-acupuncture with Neural Stem Cells Transplantation on Morphology Changes of Brain Tissue of Cerebral Palsy Rats

摘要 目的:通过透射电镜观察头部电针刺激协同神经干细胞移植对脑瘫大鼠脑组织形态学变化的影响。方法:改良HIE造模方法建立脑瘫大鼠模型,随机将实验大鼠分为假手术组、模型组、干细胞移值治疗组、电针治疗组、电针+干细胞治疗组,每组10只。各组分别予相应处理。分别于造模后1、2 W取各组实验大鼠处死取材,电镜观察脑组织形态学改变。结果:手术后1 W,假手术组神经元形态完整;模型组可见神经元水肿,细胞器数量明显减少,线粒体绝大部分嵴和部分膜融合或消失,粗面内质网明显扩张,游离核糖体减少,突触数量减少;干细胞移植组、电针组、电针协同干细胞治疗组表现相近,神经元水肿状态较模型组轻微、细胞器数量减少,少部分线粒体嵴断裂。手术后第2 W,假手术组无变化;模型组神经元水肿逐渐减轻,游离核糖体数目有所恢复,突触数密度有逐渐下降趋势;电针组神经元水肿减轻,细胞器数量增多,线粒体断裂减少,空泡样改变有所改善;干细胞移植组神经元水肿,细胞器数量略有增加,线粒体嵴断裂可见,空泡样改变减轻;电针+干细胞治疗组神经元水肿不明显,细胞器数量明显增多,线粒体嵴断裂减少,空泡样改变明显减轻,突触数密度有逐渐恢复的趋势。结论:头部电针刺激协同神经干细胞移植具有促进脑脑瘫大鼠脑神经修复作用。 Objective: To observe the effect of head electro - acupuncture with neural stem cells (NSCs) transplantation on brain tissue morphology changes of cerebral palsy rats. Methods: SD rats models were made by improved HIE method, the experimental rats were randomly divided into sham operation group, model group, NS Cs transplantation group, electro- acupunture group and electro -acupuncture with NSCs group, 10 rats in each group, each group was given the corresponding treatment. Rats were killed for pathologic sampling in 1 week and 2 week after operation, and the brain tissue morphology changes of cerebral palsy rats were observed with transmission electron microscope. Results: Neurons of sham operation group were complete in 1 week after operation. neurons of model group showed oedematous, the number of organelles decreased significantly, the most part of mitochondria crest and part of membrane fusion or disappeared, the rough endoplasmic reticulum swelled obvi- ously, free ribosome and synapse decreased ; neurons of NSCs, electro - acupuncture and eloectro - acupuncture with NSCs group soedematous eased and the number of organelles decreased, and only small number of mitochondrina crest fracture compared with model group. Neurons of sham operation group were ibidem in 2 week, neurons edema of model group gradually reduced, the number of free ribosome recovered, the synapse density were gradually declined; neurons edema of electro - acupuncture group got light, number of organelles increased, mico- chondri of crest fracture decreased, cavitation change improved ; neurons edema was seen in NSCs group, number of organelles increased lightly, mitochondrial crest fiacture was existed, cavitation change was reduced; neurons of electro - acupuncture with NSCs group edema slight, the number of organelles increased obviously, the mitochon-drial crest fracture reduced, cavitation change obviously improved, the synapse density were gradually restored. Conclusion: Neural stem cells transplantation and head electroacupuncture can significantly promote nerve restoration of cerebral palsy rats
