西洋参对卵巢早衰大鼠卵巢HPGDS PLA_2G_4A、PTGS_1基因表达的影响 被引量:3

摘要 目的:观察西洋参对卵巢早衰(POF)大鼠基因表达的影响。方法:成年雌性SD大鼠连续14 d腹腔注射4-乙烯基环己烯双环氧化物(VCD)80 mg/kg建立POF模型。模型复制成功后随机分为2组,西洋参组灌服西洋参2.25 g/kg;模型组灌服等容量生理盐水,每d 1次共30 d;正常对照组不做任何干预。对各组大鼠进行一般状况观察,采用电化学法检测血清E_2、FSH及LH水平,采用实时定量PCR(QRT-PCR)方法,检测各组大鼠卵巢组织HPGDS、PLA_2G_4A及PTGS_1的表达。结果:西洋参组大鼠较POF模型组体毛柔顺而整洁,活动敏捷;西洋参组大鼠血清激素水平更趋近于正常对照组;QRT-PCR实验结果显示,与POF模型组比较,西洋参组卵巢HPGDS、PLA_2G_4A及PTGS_1表达显著下调。结论:西洋参能够缓解卵巢的衰竭、降低VCD对卵巢的损伤。 Objective: To study the effects of of American Ginseng on HPGDS, PLA2G4A, PTGS1 expressions in POF rats. Methods: Adult female Sprague Dawley rats were intraperitoneally (i. p. ) injected with VCD (80 mg/kg) continuously for 14 days to prepare POF models. Then POF rats were randomly divided into 2 groups, American Ginseng group was given American Ginseng (2.25 g/kg), model group was given the equal volumes of saline , once a day for 30 days, normal control group without any intervention. General condition was observed in each group of rats, electrochemical method was used to detect the serum E2 , FSH and LH levels , Quantitative real- time PCR assay was used to detect the expressions of HPGDS, PLA2G4A and PTGSI. Re- suits: American Ginseng group rats showed body hair smooth, clean and tidy, activity of agile and stable mental state, compared with POF model group rats. Serum hormone levels in American Ginseng group were close to normal control group , compared with POF model group. QRT - PCR results showed that the expression levels of HPGDS, PLA2G4A PTGSl were significantly down regulated in ovary, compared with POF model group. Conclusion: American Ginseng can alleviate ovarian failure, reduce VCD on ovarian damage caused by VCD.
