摘要
目的研究8周有氧运动对视黄醇结合蛋白4(RBP4)诱导的胰岛素抵抗鼠骨骼肌和肝脏SOCS-3mRNA表达、SOCS-3与IR、IRS磷酸化蛋白结合形成的复合物的影响,探讨运动缓解RBP4诱导的胰岛素抵抗机制。方法RBP4腹腔注射建立胰岛素抵抗鼠模型。设8周有氧运动组(RBP4 aerobic exercise group,RA)、安静对照组(RBP4control group,RC)和正常安静对照组(Control group,C)。检测血清RBP4、胰岛素、血糖,计算胰岛素抵抗指数HOMA-IR;RT-PCR法检测SOCS-3mRNA表达;免疫共沉淀法检测SOCS-3与IR、IRS磷酸化蛋白结合形成的复合物表达。结果 (1)与C组相比,RC组小鼠血清RBP4、HOMA-IR均显著增加,骨骼肌、肝脏SOCS-3mRNA表达、SOCS-3与IR、IRS磷酸化蛋白结合形成的复合物均显著增加。(2)与RC组相比,RA组小鼠血清RBP4、HOMAIR均显著降低,骨骼肌、肝脏SOCS-3mRNA表达、SOCS-3与IR、IRS磷酸化蛋白结合形成的复合物均显著减少。结论 8周有氧运动能够显著改善RBP4诱导的胰岛素抵抗,机制涉及降低血清RBP4水平,减少高RBP4诱导的SOCS-3表达,以及SOCS-3与IR、IRS结合形成的复合物,恢复正常的胰岛素信号传导。
Objective To study effects of exercise on SOCS-3 expression and SOCS-3-IR/SOCS-3-IRS complex in RBP4 – induced insulin resistance mice,and explored the mechanism of exercise intervention in RBP4 – induced insulin resistance. Methods RBP4 – induced insulin resistance mice established by RBP4 intraperitoneal injection. Model rats divided in RBP4 aerobic exercise group(RA),RBP4 control group(RC) and control group(C). Results(1) RBP4,HOMA-IR,liver and skeletal muscle SOCS-3 expression and SOCS-3-IR/SOCS-3-IRS complex increased more in group FC than group C,respectively.(2) RBP4,HOMA-IR,liver and skeletal muscle SOCS-3 expression and SOCS-3-IR/SOCS-3-IRS complex decreased more in group RA than group FC,respectively. Conclusions 8-weeks aerobic exercise could decrease HOMA-IR in RBP4-induced insulin resistance mice,improve insulin resistance through decreasing liver and skeletal muscle SOCS-3 expression and SOCS-3-IR/SOCS-3-IRS complex.
