miR-145和miR-497在宫颈癌SiHa细胞中的表达及对增殖的影响 被引量:3

摘要 目的研究miR-145和miR-497在宫颈癌Si Ha细胞的表达及对增殖的影响。方法以宫颈癌Si Ha细胞为研究对象,转染pre-miR-145和pre-miR-497,采用实时荧光定量PCR和MTT检测miR-145和miR-497在宫颈癌Si Ha细胞中的表达及对增殖的影响。结果采用实时荧光定量PCR检测miR-145和miR-497在宫颈癌Si Ha细胞中的表达,结果显示,实验组中miR-145和miR-497的表达量显著高于blank组(P=0.001),MTT结果显示转染miR-145和miR-497的Si Ha细胞其增殖活性明显受到抑制(P<0.05)。结论 miR-145和miR-497能够显著抑制宫颈癌Si Ha细胞的增殖。 Objective To evaluate the effect of miR- 145 and miR- 497 on proliferation of human cervical carcinoma cells and their expression. Methods Pre- miR- 145 and pre- miR- 497 were transfected in cervical cancer Si Ha cell,and detected their expression and effect on proliferation using real- time fluorescent quantitative PCR and MTT. Results The results showed that the experimental group in the expression of miR- 145 and miR- 497 amount was significantly higher than that of blank group( P = 0. 001),and MTT showed that miR- 145 and miR-497 can significantly suppressed the proliferation of Si Ha( P〈 0. 05). Conclusion miR- 145 and miR- 497 could inhibit the proliferation of Si Ha cells.
