摘要
目的研究北京市门头沟区流行麻疹病毒(Me V)的基因型别及特征。方法采集麻疹疑似患者咽拭子和尿液标本分离病毒,提取病毒RNA,逆转录PCR扩增病毒的N基因碳末端的634 bp的核苷酸片段,对扩增产物进行序列测定和比较分析。结果在发病的39例患者中,14例扩增到N基因片段,序列阳性率为35.9%,经过序列比对,均为H1基因型。本研究所获得的14株麻疹病毒H1基因亚型在亲缘关系进化树上与WHO推荐的H基因型代表株Hunan.China93-7/H1-H1c同属一个分支,核苷酸同源性为96.7%-97.9%,氨基酸同源性为95.5%-97.3%。结论 H1基因型麻疹病毒是门头沟区麻疹病毒的优势流行株,与北京市及全国麻疹病毒流行株无差异。
Objective To study the genotype and characteristics of measles virus found in Mentougou of Beijing. Methods Measles viruses were separated from the pharynx swabs and urine of suspected measles patients. The total RNA was isolated from the measles virus. Then the 634 bp nucleotide segment of carboxylterminal region of nucleoprotein gene were amplified by RT-PCR,the sequence of the amplified segments were determined,compared and analyzed. Results In the pathogenesis of 39 patients,gene fragment N was amplified from 14 cases,the positive rate of this sequence was 35. 9%. After sequence analysis,all of the isolated viruses were H1 genotype of measles virus. The genotype H1 of measles virus which we got and the World Health Organization( WHO) H genotype representative strains Hunan. China93- 7 / H1- H1 c belong to a branch of the same tree,moreover,the homology of nucleo tide and amino acid were among 96. 7%- 97. 9% and 95. 5%- 97. 3%. Conclusion H1 genotype remains predominant in measles virus in Mentougou district,which is consistent with the strain of measles virus circulating in Beijing and the national.
