通管丸对SOI模型大鼠TNF-α的影响

Effect of Tongguan pills on TNF- α level in rats with obstructive infertility due to salpingitis

目的:探讨通管丸对炎性阻塞性不孕大鼠输卵管上皮组织中TNF-α含量的影响。方法:将84只SPF级wistar雌性大鼠随机分为正常组12只、模型组60只、假手术组12只,用混合菌接种法注入输卵管复制炎性阻塞性模型,造模成功后的大鼠随机分为通管丸高、中、低剂量组,模型对照组及妇科千金胶囊组,每组各10只。造模20d后,每组用相应的药物连续灌胃20d,第21天处死、取材,采用HE染色法观察输卵管组织形态学改变,采用免疫组化法检测输卵管上皮组织中TNF-α含量。结果:模型对照组输卵管上皮细胞TNF-α阳性表达明显高于其他各组,差异具有统计学意义(P<0.05);通管丸高剂量组TNF-α阳性表达明显低于中、低剂量组及妇科千金胶囊组,差异具有统计学意义(P<0.05);中、低剂量组与妇科千金胶囊组无明显差异(P>0.05)。结论:通管丸能够调节输卵管上皮细胞TNF-α的表达,达到消除炎症的目的,是治疗输卵管炎性阻塞性不孕的有效丸剂。

Objective： To investigate the effect of Tongguan pills on the level of tumor necrosis factor- α（ TNF-α） in the oviductal epithelial tissue of rats with obstructive infertility due to salpingitis and to provide a basis for clinical medication. Methods： Eighty- four specific pathogen- free female Wistar rats were randomly divided into normal group（ n = 12）,model group（ n = 60）,and sham- operation group（ n = 12）. For rats in the model group,a rat model of salpingitis and obstruction was established by injecting mixed bacteria into the fallopian tube. The model rats（ n = 50） were randomly divided into high-,middle-,and low- dose Tongguan pill groups,model control group,and Fukeqianjin capsule group,with 10 rats in each group. Twenty days after the model was established,the high-,middle-,and low- dose Tongguan pill groups,model control group,and Fukeqianjin capsule group received respective treatments by gavage for 20 d. These rats were sacrificed on day 21 to collect fallopian tube samples. The histomorphological changes of these samples were observed by HE staining,and the level of TNF- α in oviductal epithelial tissue was measured by immunohistochemistry. Results： The model control group had significantly higher expression of TNF- α in oviductal epithelial cells than the other groups（ P〈0. 05）. The high- dose Tongguan pill group had significantly lower expression of TNF- α than the middle- and low- dose Tongguan pill groups and the Fukeqianjin capsule group（ P〈0. 05）. There were no significant differences in the expression of TNF- α between the middle- and low- dose Tongguan pill group and the Fukeqianjin capsule group（ P〈0. 05）.Conclusion： Tongguan pills can reduce the expression of TNF- α in oviductal epithelial cells to eliminate inflammation. It is effective pills for treating obstructive infertility due to salpingitis.
