硫辛酸对帕金森大鼠黑质多巴胺能神经元的保护作用 被引量:3

PROTECTIVE EFFECTS OF ALPHA LIPOIC ACID ON THE DOPAMINERGIC NEURONS IN THE SUBSTANTIA NIGRA OF ANIMAL MODEL OF PARKINSON'S DISEASE

摘要 目的探讨硫辛酸(LA)对帕金森大鼠黑质多巴胺能神经细胞存活及炎症介质的影响。方法选用健康老年雄性SD30只,随机分为对照组、损伤组和治疗组;采用立体定向技术向损伤组和治疗组大鼠单侧黑质内注入鱼藤酮建立帕金森损伤模型,治疗组同时腹部注射LA;采用免疫组化法观察黑质酪氨酸羟化酶(TH)的表达,ELISA法检测黑质TNF-α、IL-1β炎症介质含量变化。结果与对照组比较,损伤组TH阳性细胞率和灰度值显著降低(P<0.05),治疗组高于损伤组,低于对照组(P<0.05);损伤组TNF-α和IL-1β含量减少(P<0.05),治疗组增高,但仍低于对照组(P<0.05)。结论 LA对鱼藤酮损伤老年大鼠DA能神经元细胞具有神经保护功能。 Objective To investigate the neuroprotective effects of Lipoic acid(LA)on the Dopaminergic(DA)neurons in the substantia Nigra(SNpc)of Rotenone(RT)induced animal model of Parkinson's disease(PD).Methods Aged rats received unilateral Rotenone injection into SNpc with stereotactic technique to establish the animal model,at the same time,the animal also received intraperitoneal(i.p.)injections of LA.The number of Tyrosine hydroxylase(TH)immunopositive neurons was counted using immunohistochemistry.The level of TNFαand IL-1βcontent was detected with ELISA.Results After LA treatment,the number of TH immunopositive neurons was significantly increased than that of Rotenone group,and the level of TNFαand IL-1βcontent was also reduced(P〈0.05).Conclusion LA has neuroprotective effects on the DA neurons in the Rotenone induced animal model of PD.
