组蛋白去乙酰化酶抑制剂Trichostatin A对细胞周期素D1调控的影响

摘要 目的观察组蛋白去乙酰化酶抑制剂Trichostatin A(TSA)对细胞周期素D1表达的影响,探讨TSA抑制细胞周期素D1表达的分子机制。方法以不同浓度(20、100、400 nmol/L)的TSA处理A549细胞24 h,以RT-PCR、Western blot等方法检测细胞周期素D1 mRNA和蛋白的表达水平;以染色质免疫沉淀技术分析400nmol/L的TSA处理A549细胞24 h后,细胞周期素D1基因核心启动子组蛋白3第9位赖氨酸乙酰化(AcH3k9)水平。结果随着TSA处理浓度的增加细胞周期素D1 mRNA和蛋白表达水平下降。在400 nmol/L TSA作用下,细胞周期素D1启动子区组蛋白乙酰化程度下降。结论 TSA造成细胞周期素D1表达下调可能与细胞周期素D1启动子乙酰化程度下降有关。
