摘要
【目的】探讨黄柏水提物与抗菌药联用对产超广谱β-内酰胺酶(ESBLs)大肠杆菌的抑制效果,为黄柏的综合开发利用提供参考依据。【方法】采用二倍微量肉汤稀释法测定黄柏水提物与抗菌药的最小抑菌浓度(MIC),并采用微量棋盘稀释法测定黄柏水提物与抗菌药联用后的分级抑菌浓度指数(FICI)。【结果】黄柏水提物对产ESBLs大肠杆菌的MIC为500 mg/m L,黄柏水提物与头孢曲松钠、阿莫西林、痢菌净、磺胺间甲氧嘧啶、头孢噻呋钠、头孢他啶、氟苯尼考、环丙沙星、诺氟沙星、左旋氧氟沙星、克林沙星、加替沙星、头孢噻肟联用后的FICI为0.50~2.01,其中,黄柏水提物与阿莫西林、头孢曲松钠、头孢噻呋钠、头孢他啶、头孢噻肟、痢菌净联用存在相加作用,与氟苯尼考联用存在协同作用,与诺氟沙星、左旋氧氟沙星、克林沙星、磺胺间甲氧嘧啶间为无关作用,与环丙沙星、加替沙星联用呈拮抗作用。【结论】黄柏水提物与青霉素类、第三代头孢菌素、痢菌净、氟苯尼考联用是通过相加或协同作用提高药物对产ESBLs大肠杆菌的抑制效果,但不宜与喹诺酮类抗菌药联合用药。
【Objective】The present experiment was conducted to investigate antibacterial effects of Phellodendron amurense Rupr water extracts combined with antimicrobials on ESBLs-producing Escherichia coli, in order to provide reference for comprehensive exploitation and utilization of P. amurense. 【Method】The minimal inhibitory concentration(MIC)of P. amurense water extracts combined with antibacterial agents were determined by double broth microdilution method.And the fractional inhibitory concentration index(FICI) of P. amurense water extracts combined with antibacterial agents to were determined by checkerboard microdilution method. 【Result】The MIC of P. amurense water extracts was 500 mg/m L.The FICI of P. amurense water extracts combined with ceftriaxone sodium, amoxicillin, mequindox, sulfamonomethoxine,ceftiofur sodium, ceftazidime, florfenicol, ciprofloxacin, norfloxacin, levofloxacin, clinafloxacin, gatifloxacin and cefotaxime ranged from 0.50 to 2.01. There was additive effect between P. amurense water extracts and any of 6 antimicrobials viz., amoxicillin, ceftriaxone sodium, ceftiofur sodium, ceftazidime, cefotaxime and mequindox, and there was synergistic effect between it and florfenicol, meanwhile, there was unrelated effect between it and any of 4 antimicrobials viz., norfloxacin, levofloxacin, clinafloxacin and sulfamonomethoxine, and there was antagonistic effect between it and either of ciprofloxacin and gatifloxacin. 【Conclusion 】P. amurense water extracts combined with penicillins, the third generation cephalosporin, mequindox and florfenicol can improve inhibitory effect on ESBLs-producing E. coli based on their additive or synergistic effect. However, combination with quinolone prove to be infeasible.
