干热河谷地区枇杷叶愈伤组织的诱导研究

摘要 以枇杷中等成熟叶片为材料,以污染率为指标对不同的无菌方法进行比较,以获得较优的灭菌处理条件;将适量的活性炭、维生素C、生长素和细胞分裂素加入MS基本生长培养基中,以获得较好的愈伤组织诱导条件。结果表明:利用75%乙醇摇动杀菌20秒和0.1%升汞摇动杀菌5分钟的处理方法,可有效控制污染。将活性炭和维生素C加入基本培养基中,以及采取暗启动培养,对缓解植物材料褐化具有一定作用。2,4-D和6-BA的使用浓度在1-1.5mg/L时均能诱导出较多的愈伤组织,最优培养基条件为MS+2,4-D1mg/L+6-BA1.5mg/L+AC3g/L+Vc100mg/L,出愈率达88%。 The study was carried out to examine the optimum condition of callus induction by using loguat leaves as explants.The optimum sterilization condition was obtained by comparing different aseptic processing,which used the pollutionrate as evaluation index.The callus was induced with MS basic medium supplemented with vitamin C( Vc),activated carbon( AC),auxin and cytokinin.The optimum induction medium for callus from loguat leaves was MS medium supplemented with Vc 100 mg / L,AC 3g / L,2,4-D1 mg / L and 6-BA1.5mg / L,which could lead a high inducing rate of 88%.Thepollutioncan be effectively controlled by using 75% alcohol for 20 seconds and 0.1% HgCl2 for 5 minutes in aseptic processing.Supplementing with Vc and AC in medium and beingcultured in dark conditions might play active roles in inhabiting the explants browning.
