摘要
采用乙醇回流法提取葛根总黄酮,通过正交试验优化提取工艺,确定最佳提取工艺为提取温度70℃,提取时间2.5 h,乙醇体积分数80%,料液比1∶20。通过α-葡萄糖苷酶活性的抑制试验测定体外降糖性能,葛根总黄酮对α-葡萄糖苷酶活性抑制率为66.7%。采用四氧嘧啶糖尿病小鼠模型,经不同剂量组葛根总黄酮连续灌胃4周测定空腹血糖值和糖耐量,结果表明葛根总黄酮低、中、高剂量都可显著(p<0.05)降低糖尿病小鼠空腹血糖含量,降糖率分别为16.1%,25.6%,23.9%;各给药组小鼠血糖值在0,0.5,1,2 h时均低于模型对照组小鼠,与模型对照组相比均能显著(p<0.05)抑制糖尿病小鼠餐后血糖的升高,改善糖耐量。
Ethanol reflux method, orthogonal test are used to optimize the extraction technology of total flavonoids from Pueraria. The optimum extraction of Pueraria total flavonoids: extraction temperature 70 ℃, extraction time 2.5 h, 80%ethanol concentration,material liquid than 1∶20. Hypoglycemic lowering effect is measured in vitro by the inhibitory to the activity of α- Glucosidase. The inhibition rate of Pueraria total flavonoids on the activity of α- Glucosidase is 66.7%,alloxan diabetic mice as model treated with different dose Pueraria total flavonoids for 4 weeks, then determining the blood glucose values and tolerance. The results of the low,medium and high doses total flavonoids of Pueraria all can be significantly(P〈0.05) reduced fasting blood glucose levels in diabetic mice,hypoglycemic rate respectively 16.1%, 25.6%, 23.9%, each treatment group blood glucose level of diabetic mice in 0,0.5,1,2 h are lower than the model group,they can inhibit the increase of postprandial hyperglycemia in diabetic mice compared to the model group, improveing glucose tolerance, the difference is significant(P〈0.05).
