高甘油三酯血症相关性急性胰腺炎模型的建立研究

STUDY ON THE MODEL OFHYPERTRIGLYCERIDEMIA-ASSOCIATED ACUTE PANCREATITIS

目的:探讨高甘油三酯血症相关性急性胰腺炎(HTGP)模型的建立。方法:将42只雄性SD大鼠随机分为A组(12只)、B组(30只),分别用普通饲料、高脂饲料饲养。满4周时行眶后静脉丛取血,测大鼠血清甘油三酯(TG)含量。A组随机分为2个亚组:AP组和C组,分别用雨蛙肽50μg/kg腹腔注射干预及生理盐水作为对照。B组也随机分为5个亚组:HTG+AP1组、HTG+AP2组、HTG+AP3组、HTG+AP4组和HTG组,分别用雨蛙肽50μg/kg、40μg/kg、30μg/kg、20μg/kg腹腔注射和生理盐水干预;苏木素—伊红染色(HE)光镜下观察胰腺病理变化,用底物酶法测定大鼠血清淀粉酶(AMY)含量。结果:经高脂饲料饲养后B组大鼠TG为(3.11±0.73)mmol/L,较A组的(0.99±0.30)mmol/L显著升高(P<0.01);与AP组相比,HTG+AP1组、HTG+AP2组胰腺病理评分、血清AMY含量显著升高(P<0.05),但HTG+AP3组、HTG+AP4组与AP组相比差异无统计学意义(P>0.05)。结论:高甘油三酯血症能够加重雨蛙肽诱导的急性胰腺炎的胰腺损伤。

Objective.To establish hyp otis. Forty-two male Sprague Dawley （normal fat diet group, 12 rats） and weeks. The blood sample was obtain （TG） level. Group A was randomly ertriglyceridemia-associated acute pancreatitis （HTGP） model. Methods were selected and randomly divided into two groups. Group A Group B （high fat diet group,30 rats）. Likewise, and C group （equal saline as Group B was randomly divided into five subgroups （six rats/group）, i.e. HTG＋ API group （50 μg/kg caerulein treatment）, HTG＋ AP2 group （40 μg/kg caerulein treatment）, HTG＋AP3 group （30μg/kg caerulein treatment）, HTG＋AP4 group （20 μg/kg caerulein treatment）, and HTG group （equal saline treatment）. Pancreas was carefully removed for microscopic examination with Hematoxylin-Eosin Stain. The serum amylase was assayed in different groups. Results：Compared with Group A, serum TG levels in Group B （3.11±0.73） mmol/L increased significantly （0.99±0.30） mmol/L （ P 〈 0.01）. Compared with the AP group, histological score of the pancreas and serum amylase level in the HTG＋AP1, HTG＋AP2 group increased significantly （ P 〈0.05）. No difference was observed in HTG＋ AP3, HTG＋AP4 group （ P 〉0.05）. Conclusion： Hypertriglyceridemia could accelerate the severity of caerulein-induced acute pancreatitis.
