从江香猪BMP15基因多态性研究及单倍型构建 被引量:5

摘要 研究BMP15基因多态性及单倍型与从江香猪繁殖性状的相关性,寻找影响从江香猪繁殖性状的QTL,为分子标记辅助选择提供依据。以从江香猪母猪为研究对象,利用DNA直接测序技术,对BMP15基因多态位点进行筛选和基因型分析,采用SHEsis在线软件构建单倍型。结果表明:在受试群体中,发现BMP15基因存在3个SNPs位点,其中C122T位于exon1,T111C位于exon2,A13G位于intron1;C122T和T111C突变均为错义突变,分别导致半胱氨酸(Cys)变为精氨酸(Arg),组氨酸(His)变为酪氨酸(Tyr)。基于上述3个SNPs位点,构建了6种单倍型,其中H1和H3为主要单倍型,频率分别为64.2%、24.3%;最小二乘分析发现,H3单倍型在初产和经产母猪中,总产仔数、产活仔数、乳头数均为最高,但差异性均不显著(P>0.05);C122T位点与初产母猪的总产仔数、产活仔数、乳头数显著相关(P<0.05),而T111C位点与初产和经产母猪的总产仔数、产活仔数、乳头数差异性均不显著(P>0.05),但CC基因型和EE基因型均为最多;在A13G位点上,经产母猪AB基因型总产仔数、产活仔数、乳头数均比AA基因型多,且差异显著(P<0.05)。可以考虑将H3单倍型和CC基因型、AB基因型及EE基因型作为影响从江香猪产仔数和母猪乳头数的有利单倍型和基因型。 To research the correlation between polymorphisms an d haplotypes of BMP15 gene and reproductive traits for Congjiang pigs, and look for QTL that affected reproductive traits of Congjiang pigs, providing basis for molecular marker-assisted selection, which Congjiang pigs were served as research objects, using DNA direct sequencing technology to detect polymorphic sites and analyze genotype, online software SHEsis was used to build haplotypes.There were 3 SNPs sites found in BMP15 gene for the test group, which C122 T located exon1, T111 C located exon2,A13 G located intron1. The C122 T and T111 C were missense mutations, which respectively resulted in cysteine(Cys)into arginine(Arg), histidine(His) into tyrosine(Tyr). Based on the 3 SNPs, 6 haplotypes were constructed, which H1 and H3 were the main haplotype that frequencies respectively were 64.2%, 24.3%. The least squares analysis showed that H3 haplotype was the highest among primiparous and multiparous sows for the total number born(TNB), numberborn alive(NBA)and teats, but the difference was not significant( P〉0.05). The C122 T sites was significantly correlated with TNB, NBA and teats(P 〈0.05), and the T111 C sites was not significant difference with TNB, NBA and teats of primiparous and multiparous sows(P〉0.05), but CC genotype and EE genotype sows were all maximum. For the A13 G site, AB genotype was more than AA genotype for TNB, NBA and teats of multiparous sows. The H3 haplotype and CC genotype, AB genotype and EE genotype were regarded as favorable haplotype and genotypes that affected number born and teats of Congjiang pig sows.
