UPLC测定红毛五加嫩叶中的绿原酸 被引量:3

Determination of chlorogenic acid from Acanthopanacis giraldii tender leaves by UPLC

摘要 目的采用UPLC测定红毛五加嫩叶试制品中绿原酸的含量并比较差异。方法 Acquity UPLCBEH C18柱(50 mm×2.1 mm,1.7μm),柱温30℃,流动相为乙腈-2‰磷酸溶液(10∶90),检测波长327 nm,流速0.2 m L·min^-1。结果绿原酸0.02~0.72 mg·m L^-1与峰面积具有良好的线性关系,回归方程为:Y=3×10^7X-9.9481×10^4(r2=0.9997,n=9),平均回收率为97.7%,RSD=0.61%(n=6),红毛五加嫩叶晒干品中绿原酸的含量最高(0.0326 g·g^-1),百合粉中绿原酸含量为0。结论所用方法简便、准确、重复性好,能为红毛五加嫩叶的开发提供参考。 OBJECTIVE To establish a UPLC method to determine the content of chlorgenic acid from Acanthopanacis giraldii tender leaves and compare the different contents. METHODS The analysis was carried out at 30 ℃ on an ACQUITY UPLC BEH C18( 50 mm × 2. 1 mm,1. 7 μm) column eluted with a mobile phase consisted of acetonitrile( A)- water with 2‰ phosphoric acid( B)( 10∶90). The detective wavelength was 327 nm with the flow rate of 0. 2 m L·min^-1. RESULTS The calibration curve of chlorogenic acid was linear within the concentration range of 0. 02 mg·m L^-1to 0. 72 mg·m L^-1with r2 of 0. 9997( n = 9). The average recovery was97. 7% with the RSD of 0. 61%( n = 6). The tender leaves dried by sun contained the highest content of chlorogenic acid and per gram of crude drug contained 0. 0326 g of chlorogenic acid. CONCLUSION This method is simple,accurate and repeatable. The experi-mental results can provide useful data for the development of tender leaves products.
