摘要
通过给免疫抑制小鼠灌服不同剂量的灵芝多糖,考察其对免疫抑小鼠的免疫调节及抗氧化功能的影响。将受试小鼠随机分为空白对照组(Ⅰ组)、试验对照组(Ⅱ组)、灵芝多糖低剂量组(Ⅲ组)、灵芝多糖中剂量组(Ⅳ组)、灵芝多糖高剂量组(Ⅴ组),连续给药7 d。通过脾脏或胸腺(mg)与体质量(g)的比值法检测各组小鼠脾脏和胸腺指数;以BC-ELISA法检测血清中IL-2、TNF-α的含量;采用紫外分光光度法检测肝组织中谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)、过氧化氢酶(CAT)及丙二醛(MDA)的含量。结果表明,试验Ⅱ组与Ⅰ组相比,脾脏和胸腺指数极显著降低,血清中IL-2、TNF-α的含量及肝组织中GSH-Px、SOD、T-AOC、CAT的含量均极显著减少,但MDA含量极显著增加。而Ⅲ、Ⅳ、Ⅴ组与Ⅱ组相比,各剂量组均能显著提高小鼠脾脏及胸腺指数,增加小鼠血清中IL-2、TNF-α的含量,并提高GSH-Px、SOD、T-AOC、CAT水平和降低MDA的含量。结果提示灵芝多糖各剂量组均可提高免疫抑制小鼠的抗氧化和免疫调节功能。
Different doses of polysaccharides from Ganoderma Lucidum were given to the immunosuppressive mice to study the effects of polysaccharides on immune modulation and antioxidation function of mice. Fifty mice were randomly divided into blank control group(groupⅠ), experimental control group(group Ⅱ), low dose of polysaccharide group(group Ⅲ), medium dose of polysaccharide group(group Ⅳ), high dose of polysaccharide group(groupⅤ), and Ganoderma Lucidum polysaccharides were continuously administrated to mice for 7 days. The spleen and thymus indexes were tested by the ratio of the spleen or thymus(mg)to bodyweight(g), the concentrations of IL-2 and TNF-α in serum were assayed by ABC-ELISA, and the levels of liver glutathione peroxidase(GSH-Px), super-oxide dismutase(SOD), total antioxidant capacity(T-AOC), catalase(CAT), and maleic dialdehyd(MDA) were detected by using ultraviolet spectrophotometer. Results: Compared with group Ⅰ, the spleen and thymus indexes were very significantly decreased, the concentrations of IL-2 and TNF-α in serum and the levels of liver GSH-Px, SOD, T-AOC and CAT were very significantly decreased, but the level of liver MDA was very significantly increased in group Ⅱ. And compared withⅡ group, the spleen and thymus indexes, the levels of IL-2 and TNF-α in serum and the levels of liver GSH-Px, SOD, T-AOC and CAT were all significantly improved, but the level of liver MDA was decreased in group Ⅲ, Ⅳ andⅤ. Conclusions: Ganoderma Lucidum polysaccharides can enhance antioxidant and immune modulation function in immunosuppressed mice.
