芪连扶正胶囊对乳腺癌MCF-7细胞增殖及B7-CD28共刺激通路的影响

Effect of Qilian Fuzheng Capsule on MCF-7 Cell Proliferation and B7-CD28 Costimulatory Pathway of Breast Cancer

目的:研究芪连扶正胶囊对乳腺癌MCF-7细胞增殖的影响及对B7-CD28共刺激通路的调控作用。方法 :常规培养MCF-7细胞,按实验分组干预,运用MTT法检测各组细胞增殖情况,应用流式细胞仪检测各组细胞表面B71、B72的表达。结果 :各浓度芪连扶正胶囊含药血清均能抑制乳腺癌MCF-7细胞增殖,用药浓度增加则抑制作用增强;各浓度芪连扶正胶囊含药血清组MCF-7细胞株表面B71、B72表达均高于对照组,差异有统计学意义(P<0.05),与阿霉素(ADM)合用时作用增强。结论:芪连扶正胶囊能抑制乳腺癌细胞增殖及促进共刺激分子B71、B72的表达,与阿霉素合用时作用增强。

Objective：To study the effect of Qilian Fuzheng capsule on MCF-7 cell proliferation of Breast cancer and the regulation of B7-CD28 costimulatory pathway. Methods：The MCF-7 cell line was normally cultivated and differently intervened in different experimental group. The MTT was performed to detect the cell proliferation of each group and flow cytometry was used to determine the expression of B71 and B72 of cells in each group. Results：Serums containing Qilian Fuzheng capsule of different dose all had inhibition on proliferation of MCF-7 cell and the inhibition was enhanced when the drug dose increased. Compared with the control group,serums containing Qilian Fuzheng capsule of different dose all can promote the expression of B71 and B72 significantly and the difference was statistically siginificant（P〈0.05）. The effect would increase when ADM is used together. Conclusion：Qilian Fuzheng capsule can inhibite the cell proliferation of breast cancer and promote the expression of B71 and B72. And the effect would increase when ADM is used together.
