丹参对子宫内膜异位症间质细胞MMP-9mRNA及蛋白表达的影响 被引量:3

Effect of Salviae Miltiorrhizae on Stromal Cells MMP-9 m RNA and Protein Expression of Endometriosis

摘要 目的:观察丹参对子宫内膜异位症间质细胞基质金属蛋白酶9(MMP-9)m RNA和蛋白表达的影响,探讨其可能的机制。方法:用混合酶消化法进行间质细胞培养,分别加入空白对照(空白组),不同浓度丹参(1 mg/L,5 mg/L,10 mg/L,15 mg/L)(丹参1组、丹参2组、丹参3组、丹参4组)。细胞常规培养48 h后,用CCK-8法观察不同浓度的丹参对间质细胞增殖的影响,RT-PCR法检测其对MMP-9 m RNA表达的影响,Western blotting检测其对MMP-9蛋白含量的影响。结果:CCK-8法结果显示,除丹参1组外,其余丹参组间质细胞增殖与空白组比较,差异均有统计学意义(P<0.05)。与空白组比较,除丹参1组外,各丹参组MMP-9m RNA表达降低、MMP-9蛋白表达量减少,差异均有统计学意义(P<0.05)。结论:丹参可以抑制异位内膜间质细胞增殖,同时可以抑制MMP-9m RNA和蛋白的表达,从而降低对细胞外基质的降解,最终抑制子宫异位内膜的种植、侵袭和转移。 Objective:To observe the effect of salviae miltiorrhizae on matrix metalloproteinase 9(MMP- 9) m RNA and protein expression of endometriosis and discuss their feasible mechanism. Methods:Cultured stromal cells by the method of mixed enzymatic digestion,added respectively nothing(blank group)or different concentration of salviae miltiorrhizae(1 mg/L,5 mg/L,10 mg/L,15 mg/L)(salviae miltiorrhizae 1 group,salviae miltiorrhizae 2 group,salviae miltiorrhizae 3 group,salviae miltiorrhizae 4 group). After cells were routine cultured for 48 h, observed the effect of different concentration of salviae miltiorrhizae on stromal cell proliferation by the method of CCK- 8,and detected the effect on MMP- 9 m RNA expression by the method of RT- PCR,then detected the effect on MMP- 9 protein level by the method of Western blotting. Results:The result of CCK- 8 showed that,differences of stromal cell proliferation between blank group and salviae miltiorrhizae groups other than salviae miltiorrhizae 1 group were statistical significant(P 〈0.05). Comparing with blank group, MMP- 9m RNA expression was decreased and MMP- 9 protein expression also was reduced in salviae miltiorrhizae groups other than salviae miltiorrhizae 1 group, differences being statistical significant(P 〈0.05). Conclusion: Salviae miltiorrhizae can inhibit ectopic stromal cell proliferation,meanwhile inhibit MMP- 9m RNA and protein expression,consequently reduce degradation of extra cellular matrix,and ultimately inhibit cultivation,invasion and metastasis of uterus ectopic endometrium.
