摘要
目的探讨Hes1和Jagged1在胃癌中的表达及临床意义。方法应用免疫组织化学SP-9000法及实时荧光定量(qRT-PCR)法检测26例胃癌组织和20例癌旁正常组织中Hes1和Jagged1的表达水平及相应Hes1mRNA和Jagged1mRNA的表达情况,分析其与胃癌各临床因素之间的关系。结果 Hes1在胃癌组织中的阳性表达率(57.7%)高于癌旁正常组织(20.0%),差异有显著性(χ^2=5.161,P=0.023),而Jagged1在胃癌组织中的阳性表达率(34.6%)低于癌旁正常组织(80.0%),差异有显著性(χ^2=7.645,P=0.006)。胃癌中Hes1mRNA的表达量(1.12±0.11)明显高于癌旁正常组织(0.11±0.02),而癌旁正常组织中Jagged1mRNA的表达量(4.76±1.92)明显高于胃癌组织(1.02±0.09)。Hes1的表达与胃癌的浸润深度、分化程度、脉管浸润密切相关(P〈0.05),与年龄、性别、临床分期及有无淋巴结转移无关(P〉0.05)。Jagged1的表达与胃癌的浸润深度、分化程度、脉管浸润密切相关(P〈0.05),与年龄、性别、临床分期及有无淋巴结转移无关(P〉0.05)。胃癌组织中Hes1和Jagged1两者的表达呈负相关(r=-0.862,P=0.000)。结论 Hesl和Jagged1的异常表达与胃癌的发生、浸润及转移密切相关,检测两种蛋白及mRNA定量对于胃癌的早期诊断及预后具有一定的参考价值。
Objective To explore the expression and clinical significance of Hes1 and Jagged1 in gastric cancer. Methods Immunohistochemistry SP-9000 method and real-time quantitative( qRT-PCR) were used to detect the expression of Hes1 and Jagged1 in 26 gastric cancer and 20 cases of adjacent normal gastric tissues,and the expression of corresponding Hes1 mRNA and Jagged1 mRNA,then to analyze the relationships with all clinicopathological parameters.Results The Hes1 positive expression rate( 57. 7%) in gastric cancer was higher than that in the adjacent normal tissues( 20. 0%),the difference was statistically significant( χ^2= 5. 161,P = 0. 023),while the Jagged1 positive expression rate in gastric tissue( 34. 6%) was lower than that of the adjacent normal tissues( 80. 0%),and the difference was statistically significant( χ^2= 7. 645,P = 0. 006). Hes1 mRNA expression levels( 1. 12 ± 0. 11) in gastric cancer were significantly higher than those in adjacent normal tissues( 0. 11 ± 0. 02). Jagged1 mRNA expression levels( 4. 76 ± 1. 92) in adjacent normal tissues were significantly higher than those in gastric cancer tissues( 1. 02 ± 0. 09). The Hes1 expression of gastric cancer was closely related with the degree of invasion,differentiation,and lymphovascular invasion( P〈0. 05),but it was unrelated with age,sex,clinical stage or lymph node metastasis( P〉0. 05). Jagged1 expression was closely associated with the degree of invasion clinical stage,degree of differentiation,and lymphovascular invasion( P〈0. 05),while unasscciated with age,sex,clinical stage or lymph node metastasis( P〉0. 05). The expression of Hes1 and Jagged1 in gastric carcinoma were significantly correlated( r =- 0. 862,P = 0. 000). Conclusion The abnormal expressions of Hes1 and Jagged1 were closely related to the occurrence,invasion and etastasis of gastric carcinoma. Both protein and mRNA quantitative detection had a certain reference value for early diagnosis and prognosis of gastric cancer.
