摘要
目的优化极地真菌S-3-66-E1转化北沙参中总香豆素的条件。方法采用静息细胞转化法,考察了生物转化条件(菌体密度、底物浓度、温度、pH、摇床转速、金属离子)对转化体系的影响,并通过正交实验优化工艺参数,利用HPLC检测总香豆素的转化情况。结果确定摇瓶转化的最优条件:培养温度15℃,摇床转速240r/min,菌体密度60g·L^(-1),底物浓度0.1g·L^(-1),pH值5.9,转化时间24h,在此条件下总香豆素的转化量趋近完全。结论采用静息细胞法转化北沙参中总香豆素的工艺稳定可行。
Objective To optimize the transformation conditions of total coumarins fromGlehnia littoralis by polar fungus S-3-66-E1.Methods The effects of biological transformation conditions(cell concentration,substrate concentration,temperature,pH,rotation speed,metal ion)were optimized by orthogonal design,and detected by high performance liquid chromatography(HPLC).Results The optimal transformation conditions were determined as follows:culture temperature 15℃;rotation speed 240r/min;cell concentration 60g·L-1;substrate concentration 0.1g·L-1;pH 5.9;conversion time 24 h,which the amount of total coumarins transformed completely.Conclusion The transformation condition of the resting cell of total coumarins fromGlehnia littoralis by polar fungus S-3-66-E1 was stable and feasible.
