摘要
目的:观察γ-谷维素对过氧化氢诱导人肝细胞(L02)氧化损伤的增殖和保护作用的影响。方法:建立过氧化氢氧化损伤模型,采用不同浓度的γ-谷维素对L02细胞进行处理,检测L02细胞增殖率、丙二醛和微量谷胱甘肽含量、超氧化物歧化酶和过氧化氢酶活力及活性氧水平的变化。结果:γ-谷维素浓度在0.1~0.4 mmol/L对H2O2引起的细胞损伤具有修复作用,同时降低丙二醛含量和活性氧水平,提高超氧化物歧化酶和过氧化氢活性以及谷胱甘肽含量,且呈量效关系。结论:L02细胞经0.1~0.4 mmol/Lγ-谷维素预处理后,可以缓解H2O2造成的氧化损伤,维持正常的生理形态。
Objective: To investigate the proliferative and protective effects of γ-oryzanol on L02 cells. Methods: The mode l of oxidative damage was established on L02 cells induced by H2O2. Cell viability, the contents of malondialdehyde(MDA) and glutathione(GSH), the activities of superoxide dismutase(SOD) and catalase(CAT), and the level of ROS were assayed on L02 cells treated with γ-oryzanol at various concentrations in vitro. Results: The cell viability was enhanced significantly, and the levels of ROS and MDA were decreased significantly, whereas the content of GSH and the activities of CAT and SOD were significantly increased by 0.1–0.4 mmol/L γ-oryzanol treatment after H2O2-induced oxidative damage in a dose dependent manner. Conclusion: γ-Oryzanol can effectively attenuate H2O2-induced oxidative damage at a dose of 0.1–0.4 mmol/L and maintain the normal physiology of the cells.
