角类药材组织切片方法的改进

Improvement on the methods of making tissue sections of traditional Chinese medicine used cornua

目的改进制作角类药材组织切片的技术方法。方法采用滑走切片对角类药材进行常规切片取材,用质量分数为1%的番红溶液染色,用蒸馏水冲洗,用无水乙醇-冬青油(体积比为1∶1)溶液透明,最后用加拿大树胶封藏。结果采用此方法制片并在显微镜下观察发现,切片组织结构完整、细胞形态清晰、染色效果好,药材皮层组织、间质组织以及髓腔都清晰可见。结论该方法可为角类药材的真伪鉴别及教学质量的完善提供参考。

Objective To improve the technology and methods for making tissue sections of traditional Chinese medicine used cornua. Methods Slides section method was adopted in the conventional sectioning of traditional Chinese medicine used cornua which was stained with 1%（ φ） safranine aqueous solution and rinsed with distilled water. Anhydrous ethanol-wintergreen oil（ V ∶ V = 1 ∶ 1） solution was then used to transparentize the cornua which were lastly cemented with Canadian gum. Results With the application of this method,the well-stained section remained an intact tissue structure with clear cells,cortex tissues,mesenchymal tissues and medullary cavity under microscopic observation. Conclusions The method is quick and easy to perform in making tissue sections of good quality,and it can be applied to the authentication of TCMused cornua and the improvement of teaching quality.
