摘要
目的:探讨远志对糖尿病周围神经病变(DPN)大鼠坐骨神经Bcl-2和Bax蛋白表达的影响。方法:Wistar大鼠随机分为正常组、DPN模型组和远志治疗组。DPN模型组和远志治疗组大鼠均建立DPN大鼠模型。DPN成模后,远志治疗组大鼠给予远志灌胃6周。采用HE染色法观察各组大鼠坐骨神经形态结构的变化,免疫组织化学染色法检测各组大鼠坐骨神经Bcl-2和Bax蛋白的表达情况。结果:与正常对照组大鼠比较,DPN模型组大鼠坐骨神经的形态结构发生明显病理变化;并且,Bcl-2蛋白的表达明显降低,Bax蛋白的表达明显升高(P<0.05)。与DPN模型组比较,远志治疗组大鼠坐骨神经的形态结构明显改善;并且,Bcl-2蛋白的表达明显升高,Bax蛋白的表达明显降低(P<0.05)。结论:远志可通过升高Bcl-2表达、降低Bax表达,抑制神经纤维变性,对DPN大鼠坐骨神经损伤具有保护作用。
Objective: To investigate the effects of polygala on Bcl-2 and Bax protein expression in sciatic nerve of diabetic peripheral neuropathy(DPN) rats. Methods: Wistar rats were randomly divided into normal group, DPN model group and polygala treatment group. The rats in DPN model group and polygala treatment group were made DPN rats' model. Then the rats in polygala treatment group were lavaged with polygala for 6 weeks. The morphology of sciatic nerve was observed by HE staining; The expressions of Bcl-2 and Bax protein in sciatic nerve were detected by immunohistochemical stain. Results: Compared with normal group, the morphology of sciatic nerve of rats in DPN model group changed obviously, and the Bcl-2 expression decreased while the Bax expression increased obviously(P〈0.05). Compared with DPN model group, the morphology of sciatic nerve of rats in polygala treatment group improved obviously, and the Bcl-2 expression increased while the Bax expression decreased obviously(P〈0.05). Conclusions: Polygala can inhibit nerve degeneration by increasing Bcl-2 expression and decreasing Bax expression, so it has protective effects on sciatic nerve injury of DPN rats.
