三种胶质瘤细胞株U87、U251及T98G体外趋化血液单核细胞的实验研究

Experimental Study on The Chemotaxis of Blood Monocyte to Three Kinds of Glioma Cell Lines(U87,U251 and T98G) in Vitro

摘要 目的观察3种胶质瘤细胞株(U87、U251、T98G)对血液单核细胞的趋化作用,探讨胶质瘤组织相关巨噬细胞的来源及其临床病理意义.方法应用Transwell法观察3种胶质瘤细胞株(U87、U251、T98G)对血液单核细胞的趋化作用.结果 3种胶质瘤细胞株(U87、U251、T98G)对血液单核细胞的趋化活性高于空白对照组(无血清培养液)(P<0.01);其中U87细胞株对血液单核细胞的趋化活性高于U251、T98G细胞株(P<0.01),U251细胞株对血液单核细胞的趋化活性高于T98G细胞株(P<0.05).结论 3种胶质瘤细胞株均具有趋化血液单核细胞能力,提示血液中单核细胞是胶质瘤相关巨噬细胞的重要来源,在胶质瘤发生、发展中起到重要作用. Objective To observe the chemotaxis of blood monocyte to three kinds of glioma cell lines(U87,U251 and T98G), and discuss the source of tumor associated macrophages in glioma and its clinicopathologic significance. Me thod Transwell assay was applied to observe the chemotaxis of blood monocyte to three kinds of glioma cell lines(U87, U251 and T98G). Re s ults The chemotaxis of blood monocyte to the three kinds of glioma cell lines(U87,U251 and T98G) was higher than that in the control group(P〈0.01). The chemotaxis of blood monocyte to U87 was higher than that of U251 and T98G(P=0.001). The chemotaxis of blood monocyte to U251 was higher than that of T98G(P =0.03). Conclus ion The three kinds of glioma cell lines all have the ability to chemoattract blood monocyte, suggesting that blood monocytes are one of the important sources of glioma associated macrophages,and play an important role in the occurrence or development of the glioma.
