摘要
目的检测CXCL2蛋白在结直肠癌中的表达情况,探讨其临床研究意义。方法利用Oncomine芯片数据库分析6个独立的研究中结直肠癌CXCL2mRNA的表达数据。采用组织芯片(TMA)免疫组化染色检测结直肠癌CXCL2蛋白的表达情况,并用统计学分析其与临床病理特征和预后的相关性。结果 CXCL2在结直肠癌较其癌旁组织中高表达;31.31%的结直肠癌细胞质CXCL2高表达,与肿瘤部位(P=0.022)、原发肿瘤的浸润程度(P=0.005)及淋巴结转移(P=0.017)显著相关;Kaplan-Meier和Cox回归分析表明,CXCL2高表达(P<0.001)是结直肠癌预后不良的一项独立预测因素。结论 CXCL2高表达可能与结直肠癌的侵袭和转移有关,可作为结直肠癌的预后指标和治疗的潜在靶点。
Objective To detect the expression condition of chemokine(C-X-C motif)ligand 2(CXCL2)in colorectal cancer(CRC)and to study its significance in clinical research.Methods The data of colorectal cancer CXCL2 mRNA expression in 6independent research centers were analysed by using the Oncomine microarray database.The tissue microarray(TMA)immunohistochemistry staining was employed to characterize the expression of the CXCL2 protein in CRC.The correlation of CXCL2 expression with clinicopathological features and prognosis was analyzed by statistics.Results CXCL2 was highly expressed in CRC tissues compared with corresponding peritumoral tissues.31.31% of CXCL2 cytoplasm in CRC was highly expressed,which was significantly correlated with the tumor location(P=0.022),infiltration extent of primary tumor(P=0.005)and lymph node metastasis(P=0.017).The Kaplan-Meier and Cox regression analysis revealed that high CXCL2 expression was an independent predictive factor of poor prognosis in CRC.Conclusion High CXCL2 expression may be associated with invasion and metastasis of CRC,which can be used as a prognostic index and potential target for the treatment of CRC.
