摘要
目的观察依那西普对大鼠坐骨神经结扎(CCI)模型机械性痛阈影响和肿瘤坏死因子TNF-α表达变化,探讨治疗神经病理性疼痛的可能方法。方法通过结扎大鼠一侧坐骨神经建立坐骨神经损伤的神经病理性疼痛模型,随机分为依那西普实验组和生理盐水对照组,腹腔给予1 mg依那西普或等量生理盐水对照,观察术前和术后1、3、7、14 d大鼠机械性痛阈变化即机械缩足反射阈值(MWT),采用酶联免疫法(ELISA)定量测量CCI大鼠损伤坐骨神经TNF-α表达变化。结果依那西普组CCI大鼠在第1、3和7天的损伤坐骨神经TNF-α表达明显低于对照组,在第7、14天的机械缩足反射阈值明显高于对照组(P<0.05或0.01)。结论依那西普抑制CCI大鼠损伤坐骨神经结扎TNF-α表达,减轻神经病理性疼痛模型CCI大鼠的机械性痛阈。
Objective To investigate the effect of etaneroept on mechanical hyperalgesia and expression of tumor necrosis factor- alpha in rat models with chronic constriction injury and to explore possible therapies to treat neuropathic pain. Methods The rat models with neuropathic pain caused by sciatic nerve injury were established by ligating the sciatic nerves of one side. The rat models were randomly divided into two groups,the etaneroept group and the control group of normal saline,given 1 mg of etaneroept and normal saline respectively. The paw withdrawal mechanical thresholds before operation and 1,3,7,14 days after operation were assessed and the expression changes of TNF- α in damaged sciatic nerves of the rats with CCI were observed. Results The expression of TNF- α in damaged sciatic nerves of the rats with CCI of the etaneroept group on Day 1,Day 3,Day7 after surgery was significantly lower than that of the control group,the paw withdrawal mechanical threshold on Day 7 and Day 14 of the former group remaining significantly higher than that in the latter one( P〈0. 05 or 0. 01). Conclusion The etanercept can lower the mechanical threshold of CCI rat models by decreasing the expression of TNF- α in damaged sciatic nerves.
