混合胶原型大鼠类风湿关节炎模型制备及评价 被引量:2

摘要 1目的制备并评价混合胶原型类风湿性关节炎(RA)大鼠模型。2方法将实验大鼠分为对照组、低剂量组和高剂量组,每组10只。分别对3组动物通过尾部注射生理盐水、0.1mg/mL混合胶原和0.4mg/mL混合胶原,1.0mL/只,间隔1周注射1次,共注射3次。末次注射1周后逐周观察并记录大鼠一般情况,并进行关节评分。末次注射3周后,处死大鼠,取右踝关节组织进行HE染色。3结果低剂量组和高剂量组关节腔炎细胞浸润清晰可见,且高剂量组炎细胞浸润较低剂量组严重。低剂量组和高剂量组关节评分在不同时期均高于对照组;低剂量组和高剂量组在末次注射2周和3周后关节评分均高于末次注射1周后。4结论对实验大鼠经过3次尾部注射混合胶原,成功建立RA动物模型,其组织学及临床表现与人类RA相似。 Objective RA model preparation and evaluation of mixed plastic prototype.MethodsExperimental animals were divided into 3groups:control,low-dose and high-dose groups,each group of 10.Respectively for the above 3sets of tail of the animal through the injection of saline,0.1mg/mL hybrid mixture of collagen and 0.4mg/mL,collagen,1.0mL/,interval injections a week,has injected 3times.After last injection a week-by-week to observe and record the animals in General,and make a joint score.Three weeks after the last injection,animals,organizations of right ankle HE staining.Results Low dose and high dose group joint infiltration of inflammatory cells visible,and the inflammatory cell infiltration in high dose group is more serious than that of in the lower dose group.Low-dose and high-dose collagen group joints scores were higher than those in the control group at different times;the joint scores of the low dose and high dose group in the last two weeks and three weeks were higher than a week after the last injection.Conclusion On rat tail mixed collagen injection after 3times,successful establishment of animal models of RA,the histological and clinical manifestations are similar to human RA.
