摘要
目的探讨胶质母细胞瘤中基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)活性酶及其中介体对原代培养肿瘤细胞侵袭力的影响。方法对21例胶质母细胞瘤组织进行明胶酶谱检测、原代培养及Boyden小室侵袭性实验,分析MMP-2酶原、活性酶及其中介体与原代培养肿瘤细胞侵袭能力之间的关系。结果 21例胶质母细胞瘤组织中9例出现MMP-2中介体,有中介体表达组原代培养的肿瘤细胞侵袭能力高于无中介体表达组(F=15.275,P=0.001),MMP-2活性酶的含量与各时段肿瘤细胞侵袭力均呈正相关(r均>0,P均<0.05),pro-MMP-2及中介体含量与肿瘤细胞侵袭力无相关性(P>0.05)。结论胶质母细胞瘤组织中MMP-2活性酶的含量及中介体的有无是决定肿瘤细胞侵袭能力的关键因素。
Purpose To study the influence of active MMP-2 and intermediate MMP-2 form on invasive ability of glioblastoma cells. Methods 21 cases of gfioblastoma tissue were examined by gelatin zymography, primary culture and experiment of Boyden chamber, and statistic analysis were made for the relationship between different activation forms of MMP-2 with invasive ability of glioma cells. Results Intermediate MMP-2 was detected in 9 cases of glioblastoma tissue. The transmembrane cell number of cases with intermediate MMP-2 form were more than those cases without intermediate MMP-2 ( F = 15. 275, P = 0. 001 ). There was positive relationship be- tween the counts of active MMP-2 and ability of glioma cells ( r 〉 0, P 〈 0. 05 ). The correlation analysis between pro-MMP-2 or intermediate MMP-2 form and glioma cells invasiveness had no statistical significance. Conclusions Counts of active MMP-2 and intermediate form existence are very important factors to ability of glioblastoma cells.
