甲醇-乙醇混合溶剂萃取脱酚制备脱酚棉籽蛋白的工艺研究 被引量:3

摘要 以脱脂棉籽湿粕为原料,利用甲醇-乙醇混合溶剂萃取脱酚制备脱酚棉籽蛋白,通过单因素试验和正交试验重点探讨了甲醇-乙醇混合比例、混合溶剂体积分数、萃取温度、萃取时间和料液比对脱酚棉籽蛋白游离棉酚含量和蛋白质氢氧化钾溶解度的影响。确定的最佳工艺条件为:甲醇-乙醇混合比例1∶1(质量比),甲醇-乙醇混合溶剂体积分数90%,萃取温度55℃,每次萃取时间25 min,料液比1∶3,分3次进行。在最佳工艺条件下制备的脱酚棉籽蛋白游离棉酚含量为157mg/kg,蛋白质氢氧化钾溶解度为62.7%。 Degossypolized cottonseed protein was prepared from wet defatted cottonseed meal by mixed solvent of methanol and ethanol,and the effects of proportion of methanol- ethanol mixture,volume fraction of mixed solvent,extraction temperature,extraction time and ratio of solid to liquid on the content of free gossypol and potassium hydroxide solubility of protein of degossypolized cottonseed protein were mainly investigated by single factor experiment and orthogonal experiment. The results indicated that the optimal process conditions were obtained as follows: mass ratio of methanol to ethanol in mixed solvent 1∶ 1,volume fraction of mixed solvent 90%,extraction temperature 55 ℃,extraction time 25 min every time,ratio of solid to liquid 1∶ 3,extraction for three times. Under the optimal conditions,the content of free gossypol in degossypolized cottonseed protein was 157 mg / kg,and the potassium hydroxide solubility of protein was 62. 7%.
