油用牡丹籽饼粕中低聚茋类含量测定方法研究 被引量:8

摘要 采用Folin-Ciocalteus比色法测定油用牡丹籽饼粕中低聚芪类化合物的含量。以没食子酸为标准物质,系统研究了Folin-Ciocalteus比色法测定油用牡丹籽饼粕中低聚芪类的方法。得到适宜的显色条件为:显色剂用量为5mL,碳酸钠溶液用量为15mL,反应时间为30min,反应温度为20~25℃。根据低聚芪类化合物与Folin-Ciocaheus反应后的紫外可见光区的特征吸收,由光谱扫描曲线可知最大吸收波长为750nm,通过绘制标准曲线,得到没食子酸的浓度与吸光度之间的线性关系。研究结果表明:在0.5~3.5μg/mL范围内,没食子酸吸光度与其质量呈良好线性关系,R2=0.9996,平均加样回收率99.8%,RSD为0.97%。该方法简便、快速、稳定性好,可以用于低聚芪类化合物的含量测定。 The experiment in the paper utilized Folin - Ciocalteu colorimetric method to determine the o benes in peony seed cake for oil. The gallic acid was applied as a reference substance and has been systematically investigated for the development of the optimal colorimetric method. The optimum colorimetric conditions were as follows: 5 mL of Folin -Ciocalteu reagent, 15 mL of 20% Na2CO3 solution, reaction time for 30 min and reaction tem- perature at 20 - 25 ℃. On condition that the reaction of oligostilbenes and the Folin - Ciocalteu reagent had the characteristics of absorption at 400 - 900 nm UV - Vis, the spectral scanning curves was taken to determine the wavelength as 750 nm. By drawing standard curve, the significant linear relation between concentration of gallic acid and absorbance was obtained. The calibration curve for quantifying gallic acid exhibited a fine linearity in range of 0. 5 - 3.5 μg/mL (R2 =0.999 6) ; the reaverage recoveries of gallic acid of 99.8% with a RSD of 0.97%, the results showed that the method is simple, fast, fine stability, and suitable for the determination of the oligostilbenes in the plant.
