Immobilization of Cephalosporin C Acylase

摘要 本研究采用传统的包埋法、吸附法和共价结合法固定化头孢菌素C酰化酶,以表观酶活力回收率为指标,结果显示共价结合法的固定效果较好,其中ES-1载体固定化酶的表观酶活力回收率约30.9%。优化ES-1载体固定化条件,当酶的载量为150 u/g,磷酸钾缓冲液浓度1.0 mol/L,p H 9.5,25℃轻微振荡12 h,固定化效果较好,表观酶活力为60.73 u/g,表观酶活力回收率约40.5%。 Three traditional methods of immobilization, such as entrapment, adsorption and covalent were applied for the immobilization of cephalosporin C acylase. The apparent recovery of enzyme activity was used as the index for the investigation of the optimal immobilization method. It indicated that the covalent was the most suitable method, in which the apparent recovery of ES-1 immobilized enzyme reached about 30.9%. The immobilization conditions of ES-1 were also optimized as enzyme investment amount of 150 μ/g, potassium phosphate buffer at 1.0 mol/L, pH 9.5 and shaking for 12 h at 150 r/min. The apparent activity was 60.73 μ/g, with the apparent recovery of 40.5%.
