摘要
目的研究苜蓿素对小鼠免疫功能的影响。方法用DNFB制备KM种小鼠迟发型超敏反应(DTH)模型,用打孔器取下直径8 mm的耳片,摘取脾脏和胸腺组织,称量上述组织,计算肿胀度抑制率和脾脏、胸腺指数;取正常小鼠脾脏,淋巴细胞培养,分别用刀豆蛋白、脂多糖刺激分化成T、B淋巴细胞,分别加入不同浓度(20、10、5、1、0.5和0.1μg/m L)苜蓿素,37℃,48 h后,测定吸收度并计算受试药对淋巴细胞增殖的抑制率。结果苜蓿素剂量5 mg/kg和10 mg/kg均能减轻小鼠耳肿胀程度且降低DTH小鼠的胸腺指数,10 mg/kg苜蓿素能降低其脾脏指数,但是5 mg/kg苜蓿素并没有降低DTH小鼠的脾脏指数;体外细胞实验发现:苜蓿素终浓度为20、10、5、1、0.5和0.1μg/m L时能抑制正常小鼠T淋巴细胞的增殖,苜蓿素终浓度为20、10、5、1和0.5μg/m L时能抑制正常小鼠B淋巴细胞的增殖。结论苜蓿素具有一定的免疫抑制作用。
Objective To study the effect of tricin on immune function in mice. Methods The mouse model of Delayed- type hypersensitivity( DTH) was induced by DNFB. Bolts were removed by perforator with an 8- mm diameter. Spleen and thymus tissues were also collected. The above tissues were weighed and the degree of swelling inhibition rate,as well as the spleen and thymus indices was calculated. Lymphocytes were cultured from spleen in normal mice.Con A and LPS was used to stimulate lymphocyte differentiation into T and B cells,which were then treated with different concentrations( 20、10、5、1、0. 5 and 0. 1 μg / m L) of tricin at 37℃. After 48 h,tricin absorption was detected and lymphocyte proliferation inhibition rate was calculated. Results Tricin at 5 mg / kg and 10 mg / kg could reduce the ear swelling degree and the thymus index in DTH mice. Tricin at 10 mg / kg group reduced the spleen index,but 5 mg / kg group did not reduce the spleen index in DTH mice. In vitro experiments revealed that the proliferation of T lymphocytes in normal mice was inhibited by tricin at final concentration of 20 μg / m L,10 μg / m L,5 μg / m L,1 μg / m L,0. 5 μg / m L and 0. 1μg /m L,respectively,and B lymphocyte proliferation in normal mice was inhibited by tricin at final concentration of 20 μg /m L,10 μg / m L,5 μg / m L,1 μg / m L and 0. 5 μg / m L,respectively. Conclusion Tricin showed immunosuppressive function in mice.
