摘要
为评价不同接种途径对禽腺病毒(FAdV)在鸡胚中的增殖能力及病毒的主要分布组织,本研究将FAdV分别采用卵黄囊、尿囊膜和尿囊腔3种途径接种SPF鸡胚,利用荧光定量PCR方法对FAdV在鸡胚组织中的病毒载量进行测定。结果显示,FAdV在鸡胚各组织中均有不同程度增殖。FAdV经卵黄囊途径和尿囊腔途径接种SPF鸡胚,病毒主要嗜性组织为肝脏,经尿囊膜途径接种FAdV,病毒主要嗜性组织为尿囊膜。经卵黄囊途径接种,FAdV在肝脏、尿囊膜和尿囊液中的病毒载量分别为1.38×108拷贝/mg、2.78×107拷贝/mg及1.32×104拷贝/mg。经尿囊膜途径接种,FAdV在肝脏、尿囊膜和尿囊液中的病毒载量分别为1.83×104拷贝/mg、1.08×105拷贝/mg及4.53×104拷贝/mg。经尿囊腔途径接种,FAdV在肝脏、尿囊膜和尿囊液中的病毒载量分别为1.65×105拷贝/mg、7.08×104拷贝/mg及1.26×104拷贝/mg。以上结果表明,不同接种途径对FAdV在SPF鸡胚中的增殖影响较大,其中经卵黄囊途径接种SPF鸡胚,FAdV增殖能力最强。
To evaluate the propagation capability of fowl adenovirus(FAdV) in chicken embryonated eggs, the virus was inoculated into SPF chicken embryonated eggs by three different routes, including embryo yolk sac(YA), chorioallantoic membrane(CAM) or chorioallantoic antrum(CAA), and the virus loads in various chicken embryo organ tissues were detected by real-time PCR. The results showed that the virus distributed widely in all chicken embryo tissues, and liver was the main tropism tissue of FAdV when inoculated via the YA and CAA, while the virus propagated mainly in the chorioallantoic membrane via CAM inoculation route. Particularly, inoculation via YA route, the viral loads were 1.38 ×108copoies/mg in chicken embryo liver,2.78 ×107copies/mg in the chorioallantoic membrane and 1.32 ×104copies/mg in chorioallantoic fluid, respectively. Viral load of FAdV in the chicken embryo liver, chorioallantoic membrane and chorioallantoic fluid were 1.83 ×104copies/mg, 1.08 ×105copies/mg and 4.53 ×104copies/mg via the CAM inoculation route. In the CAA inoculation route, the viral loads were 1.65 ×105copies/mg in chicken embryo liver, 7.08 ×104copies/mg in chorioallantoic membrane and 1.26 ×104copies/mg in chorioallantoic fluid, respectively. In conclusion, the propagation of FAdV was significantly affected by different inoculation route in chicken embryonated eggs. The YA inoculation route was considered to be the most appropriate way for the virus propagating in SPF chicken embryonated eggs.
