摘要 目的:对比不同质量浓度下芍药总多糖对变异链球菌、血链球菌、黏性放线菌生长、产酸及合成水不溶性多糖(water insoluble glucan,WIG)含量的影响。方法:自行提取芍药总多糖,连续稀释法测定芍药总多糖对变异链球菌、血链球菌、黏性放线菌生长的最低抑菌浓度(minimal inhibitory concentration,MIC)与最低杀菌浓度(minimal bactericidal concentration,MBC);将芍药总多糖倍比稀释为小于MIC的4个质量浓度组,以脑心浸液液体培养基为空白对照组,加入108CFU/ml的变异链球菌、血链球菌、黏性放线菌菌悬液,厌氧培养24h及48h。p H计测定p H值,计算△p H值(初始p H-终末p H)。蒽酮法测定WIG含量。结果:1芍药总多糖对变异链球菌、血链球菌、黏性放线菌生长均有抑制作用,其最低抑菌浓度变异链球菌为110.4mg/ml,血链球菌为27.6 mg/ml,黏性放线菌为13.8 mg/ml;2培养48h后,△p H值均随芍药总多糖质量浓度的增大而减小。变异链球菌实验组除1/4MIC、1/2MIC组之外,其余各组间的△p H值差异无统计学意义(P〉0.05)。血链球菌1/2MIC组同对照组间有统计学意义(P〈0.05)。黏性放线菌各组间差异无明显统计学意义(P〉0.05);3培养24h,WIG含量随芍药总多糖质量浓度的增大而减小。变异链球菌1/2MIC组及血链球菌1/4MIC及1/2MI组同对照组差异有统计学意义(P〈0.05)。结论:芍药总多糖对浮游状态下的变异链球菌、血链球菌、黏性放线菌的生长及变异链球菌、血链球菌产糖、产酸有不同程度抑制作用。 Objective To investigate the effect of polysaccharides from Paeonia sinjiangensis K.Y. Pan on the growth, acid production, glucan generation of Streptococcus mutans,Streptococcus sanguis and Actinomyces viscosus. Methods By the method of solvent extraction process, the polysaccharides was extracted from Paeonia sinjiangensis K.Y. Pan.Two-fold dilution was used to measure Minimal inhibitory concentration(MIC) and minimal bactericidal concentration(MBC) of polysaccharide from Paeonia sinjiangensis K.Y. Pan effecting on the growth of Streptococcus mutans,Streptococcus sanguis and Actinomyces viscosus.The polysaccharides from Paeonia sinjiangensis K.Y. Pan was used as the experimental group with concentrations ranging from 1/2MIC to 1/16 MIC prepared with brain heart infusion broth(BHI) liquid medium, and BHI liquid medium was used as the control group. Using p H meter measure Δp H and Anthrone measured the effect of polysaccharide on acid ogenicity of oral cariogenic bacterial and on intracellular polysaccharide. Results 1Total polysaccharides from Paeonia sinjiangensis K.Y. Pan significantly inhibited the growth of Streptococcus mutans,Streptococcus sanguis and Actinomyces viscosus, of which MIC measured by dilution experiments were, respectively,110.4mg/ml,13.8mg/ml and 27.6 mg/ml.2Total polysaccharides from Paeonia sinjiangensis K.Y. Pan inhibited acid production of Streptococcus mutans,Streptococcus sanguis, while its effect of Actinomyces viscosus, compared with the control group, had no significant difference. 3Total polysaccharides from Paeonia sinjiangensis K.Y. Pan had obvious inhibitory effect on the synthesis of intracellular polysaccharide(WIG) of Streptococcus mutans,Streptococcus sanguis, while in the experimental concentration, it had little effect on the synthesis of intracellular polysaccharide of Actinomyces viscosus. Conclusion Total polysaccharides from Paeonia sinjiangensis K.Y. Pan inhibits in vitro on growth of cariogenic bacterial,acid production and glucan generation, effectively preventing the occurrence of dental caries.
