摘要 目的:优选葛根总黄酮的纯化工艺。方法:以洗脱液pH、洗脱液浓度、洗脱液用量为自变量,葛根总黄酮吸附-洗脱率为因变量,采用星点设计-响应面法优选纯化工艺。结果:最佳纯化工艺为:pH=3.0、洗脱液浓度95%、洗脱液用量2.47 BV。葛根总黄酮的最大解吸率达到92.0%,纯化后葛根总黄酮含量达67%。结论:本工艺操作简单,稳定可行,适合于葛根总黄酮的大量制备。 Objective: To optimize purification process of flavonoids in Radix Puerariae. Methods: Selecting pH, ethanol concentration and fluent volume as independent variables, response surface methodology was used to optimize the process by the index of the fluent volume desorption rate of total flavonoids. Results: The best purification process were as follows: the fluent pH was 3.0, eluted with 2.47 BV of 95 % ethanol. The desorption rate and the content of total flavonoids were up to 92.0 % and 67 % respectively. Conclusion: The optimized purification technology was feasible and reliable. It was suitable for large-scale preparation of total flavonoids in Radix Puerariae.
