摘要
通过改变磷酸替米考星合成起始物料,改变传统的合成方法。具体措施:将培养170 h后的泰乐菌素发酵液p H调至1.5~2.0,酸化处理一定时间后加入硅藻土过滤。向澄清滤液中加入乙酸丁酯并用氢氧化钠调节p H至11左右,震荡摇匀静置分相后向有机相中加入3,5-二甲基哌啶缓慢升温至70℃,恒温滴加甲酸溶液催化合成替米考星,合成液经水提、脱色、喷雾干燥得符合药典标准的磷酸替米考星产品。
Tilmicosin synthesis starting material changes traditional synthesis method by changing the phosphate. The culture to tylosin fermentation solution after 170 h adjusted to p H 1. 5 to 2. 0,after a certain period of time with the acidizing treatment,diatomaceous earth filtration was added. Butyl acetate was added to clarified filtrate adjusted p H to about 11 with sodium hydroxide,3,5- dimethyl piperidine was added after shaked static phase to organic phase with slow heating to 70℃,dropping formic acid catalytic in constant temperature,tilmicosin was synthesized,synthetic liquid can obtain tilmicosin products in line with the phosphorylation of pharmacopoeia standards by water extraction,decolorization and spray drying.
