摘要
目的利用基因表达谱芯片技术探讨原发性肝癌(PHC)不同阶段(癌组织、癌旁组织、正常组织)差异基因的表达,进一步寻找PHC不同阶段组织中的差异表达基因。方法提取20例肝癌癌组织、癌旁组织和正常组织的总RNA,应用Agilent人类全基因组4×44K基因芯片筛选差异表达基因,采用微阵列类分析(SAS)软件对芯片图像进行分析。结果筛选出癌组织与正常组织差异基因4 175个,其中上调基因1 850个,下调基因2 325个。对筛选出的差异基因进行GO分类,主要分为催化活性、信号转导、酶活性调节、转录活性调节、蛋白运输功能、细胞生长、凋亡等功能过程。结论利用基因芯片技术可高通量地筛选出PHC不同阶段组织的差异表达基因,为进一步阐明PHC的发生机制提供实验依据。
Objective To study the related gene expression in hepatic carcinoma of different stages( cancerous tissue,paraneoplastic tissue,normal tissue) by c DNA expression profiling microarray technique,and to further seek differentially expressed genes of hepatocellular carcinoma tissue in different stages. Methods The whole RNA of 20 cases of cancerous tissue,paraneoplastic tissue,normal tissue of hepatic carcinoma was extracted. Agilent human genome 4 × 44 K gene chip was applied to screen differentially expressed genes,and microarray analysis software( SAS software) was used to analyze chip image. Results 4 175 kinds of differentially expressed genes of cancerous tissue and normal mucosa tissue were screened,among which,there were1 850 kinds of up- regulated genes and 2 325 kinds of down- regulated genes. Then through GO classification,screened differentially expressed genes mainly were divided into the following function and process: catalytic activity,signal transduction,enzymatic activity regulation,transcription activity regulation,protein transportation function,cell growth and apoptosis,and so on.Conclusion Gene chip technology can high- throughput screen differentially expressed genes of hepatocellular carcinoma in different stages,and provide experimental basis for further expounding the occurrence mechanism of hepatocellular carcinoma.
