摘要
目的分析X射线对BGC-803胃癌细胞凋亡及Survivin蛋白表达的影响。方法将BGC-803胃癌细胞分为对照组、实验组;实验组分别给予1、2、5 Gy X射线照射。流式细胞仪检测各组细胞凋亡及Survivin蛋白表达情况。结果随着X射线剂量增大,BGC-803细胞凋亡率增大;在2 Gy、5 Gy组,随X射线照射时间延长,细胞凋亡率增大,而1 Gy组无明显改变。大剂量(5 Gy)X射线引起BGC-803胃癌细胞Survivin蛋白表达增加,而小剂量X射线(1 Gy、2 Gy组)对BGC-803胃癌细胞Survivin蛋白表达无显著影响。结论 X线辐射引起BGC-803胃癌细胞凋亡具有辐射剂量、时间依赖性;大剂量X射线照射使Survivin蛋白表达增加,可能与细胞周期、DNA同源重组修复有关。
Objective To observe the effects of X- ray on apoptosis of BGC- 803 gastric cancer cells and expression of Survivin protein. Methods The BGC- 803 gastric cancer cells were divided into control group and experimental groups( radiation doses were 1、2、5 Gy,respectively). Apoptosis of BGC- 803 gastric cancer cells and expression of Survivin protein were detected by flow cytometry. Results The apoptosis rate of BGC- 803 cancer cells was increased accompanied by the increase of X- ray dose. The apoptosis rate of BGC- 803 cancer cells in 2、5 Gy groups was increased with the passage of time,but there was no chage in 1 Gy group. Low dose( 1、2 Gy) of X- ray had no effect on the expression of Survivin protein,but 5 Gy group had increased the expression. Conclusion The apoptosis of BGC- 803 cancer cells was induced by X- ray in a dose- and time- dependent manner. High dose X- ray increased the expressiong of Survivin protein. It may be associated with DNA repair by homologous recombination or cell cycle.
